Biodegradation potential assessment of microbial consortia isolated from a diesel-contaminated soil

Abstract: Diesel, toluene and naphthalene-degrading microbial consortia were isolated from a diesel-contaminated soil. The presence of catabolic genes, xylE and ndoB responsible for toluene/xylene and naphthalene biodegradation, respectively, were screened by PCR techniques in all microbial consortia. The diesel-consortium possessed both catabolic genes, the toluene-consortium only the xylE gene, while the naphthalene-consortium possessed only the ndoB gene. On the basis of these results, it was concluded that contamina… Show more

“…For the total count of bacteria, Nutrient agar was used; for fungi, Malt agar with streptomycin; for Gram-negative bacteria, McConkey agar 13 and for microorganisms which decompose hydrocarbons, mineral base medium 14 with different carbon sources: 2000 ppm diesel fuel, 375 ppm toluene or 200 ppm phenanthrene. 15 Actinomycetes, Nocardia and Pseudomonas as well as cellulose decomposing bacteria and fungi were determined using selective media. 16,17 The content of total petroleum hydrocarbons (TPH) in the soil was determined according to DIN EN 14345:2004.…”
