Biogeography, autecology, and phylogeny of Percolomonads based on newly described species

Abstract: Percolomonads (Heterolobosea) are aquatic heterotrophic flagellates frequently found in saline waters up to hypersaline environments. We isolated and cultivated seven strains of percolomonad flagellates from marine waters and sediments as well as from a hypersaline inland lake in the Atacama Desert. Morphological characterizations, comprising light and scanning electron microscopy, revealed only slight differences between the strains mainly limited to the cell shape, length of flagella, and length of the ventr… Show more

“…Our findings support their cosmopolitan distribution and show that they are also widely distributed in benthic habitats around the Azores islands at various depths. The species Lula levis and Nonamonas santamariensis were not found in two major metabarcoding datasets in our previous study [ 63 ], leading to the assumption that they are restricted in their distribution. However, the current dataset suggests that they are as well widely distributed around the Azores islands in a wide range of depths.…”

“…The samples analyzed in this study were also used for a cultivation-based approach [ 15 ], resulting in several monoclonal cultures belonging mainly to Cafeteriaceae [ 50 ] and Percolomonadida [ 63 ]. We used these groups as model groups to check whether we can find V9 sequences belonging to our cultivated strains in the metabarcoding dataset.…”

“…from Azores) each form an individual branch. This may be signs for an allopatric speciation in remote areas/islands(Hernandez-Hernandez et al, 2021) as it was postulated for percolomonads and other protist species(Arndt et al, 2020;Hohlfeld et al, 2022). Jet, genetic differences between different protist genera can widely vary depending on the genus and family.…”

“…), in saline microbial mats and hypersaline lagoons harboring biosignatures classified as diatoms, bicosoecids, and ciliates as detected by metabarcoding (Saghaï et al, 2017;Rybarski et al, 2023). Efforts to isolate and sequence the cultivable and uncultivable protist diversity from saline to hypersaline lagoons unveiled a novel endemic diversity including species from Opisthokonta (choanoflagellates), Rhizaria (Cercomonadida), Discoba (percolomonads), Stramenopiles (bicosoecids and placidids), and Alveolata (ciliates) (Schiwitza et al, 2018(Schiwitza et al, , 2019(Schiwitza et al, , 2021Arndt et al, 2020;Carduck et al, 2021;Rybarski et al, 2021Rybarski et al, , 2023Schoenle et al, 2022;Acosta et al, 2023;Hohlfeld et al, 2023).…”
