Bioinformatic analysis of eosinophil activity and its implications for model and target species

Abstract: Eosinophils are important immune cells that have been implicated in resistance to gastrointestinal nematode (GIN) infections in both naturally and experimentally infected sheep. Proteins of particular importance appear to be IgA-Fc alpha receptor (FcαRI), C-C chemokine receptor type 3 (CCR3), proteoglycan 3 (PRG3, major basic protein 2) and EPX (eosinophil peroxidase). We used known human nucleotide sequences to search the ruminant genomes, followed by translation to protein and sequence alignments to visualiz… Show more

“…The immune response of eosinophilia among induced infected sheep plays an important role to fight against the gastrointestinal nematodes, particularly H. contortus (Balic et al, 2006;Terefe et al, 2009). However, the mechanisms of eosinophil action were reported to vary between host species (Jenvey et al, 2020;Hayyan et al, 2020) and it is species-specific (Henderson et al, 2006).…”

“…As we saw in zebrafish for Embp, hMBP-2 is exclusively expressed in eosinophils 34 , while hMBP-1, although predominantly expressed in eosinophils is also expressed in basophils and mast cells, suggesting that embp may be the ortholog of hMBP-2. Interestingly, in ruminants, MBP-2 is the only MBP present 35 . Also, the relatively neutral pI of Embp suggests that the characteristically strongly basic pI of hMBP-1 may be a relatively recent evolutionary occurance.…”

“…Also, the relatively neutral pI of Embp suggests that the characteristically strongly basic pI of hMBP-1 may be a relatively recent evolutionary occurance. Indeed, the pIs of MBP orthologues vary considerably across species 34,35 . Although further studies into the biochemical and functional properties of this zebrafish orthologue will be needed, zebrafish embp will likely offer an interesting window into the evolution of MBP proteins.…”

“…White blood cell counts are directly related to the immune response of animals. Of the white blood cells, eosinophils are the most important cells and related to the protection and immune response against infections of gastrointestinal parasites (Jenvey et al 2020;Mitre and Klion 2021). Eosinophilia is a typical consequence of infection with helminths (Hendawy 2018).…”
