Piceatannol (PIC) is a natural stilbene extracted from grape skins that exhibits biological activities such as antibacterial, antitumor, and antioxidant activities. The present study was carried out to further investigate the effect of PIC on the antibacterial activity of different antibiotics and to reveal the antibacterial mechanism of PIC. We found that PIC had an inhibitory effect against Staphylococcus aureus (S. aureus); its minimum inhibitory concentration (MIC) and minimum bactericidal concentration (MBC) were 128 μg/mL and 256 μg/ mL, respectively. Additionally, we measured the fractional inhibitory concentration (FIC) of PIC combined with antibiotics via the checkerboard method. The results showed that when PIC and ciprofloxacin (CIP) were combined, they displayed a synergistic effect against S. aureus. Moreover, this synergistic effect was verified by time–kill assays. Further, the results of the membrane permeability assay and proton motive force assay revealed that PIC could enhance the sensitivity of S. aureus to CIP by dissipating the bacterial proton motive force (PMF), particularly the ∆ψ component, rather than increasing membrane permeability. PIC also inhibited bacterial adenosine triphosphate (ATP) synthesis and was less likely to induce bacterial resistance but exhibited slight hemolytic activity on mammalian erythrocytes. In summary, the combination of PIC and CIP is expected to become a new drug combination to combat S. aureus.

Ginsenosides are major bioactive compounds present in the Panax species. Ginsenosides exhibit various pharmaceutical properties, including anticancer, anti-inflammatory, antimetastatic, hypertension, and neurodegenerative disorder activities. Although several commercial products have been presented on the market, most of the current chemical processes have an unfriendly environment and a high cost of downstream processing. Compared to plant extraction, microbial production exhibits high efficiency, high selectivity, and saves time for the manufacturing of industrial products. To reach the full potential of the pharmaceutical resource of ginsenoside, a suitable microorganism has been developed as a novel approach. In this review, cell biological mechanisms in anticancer activities and the present state of research on the production of ginsenosides are summarized. Microbial hosts, including native endophytes and engineered microbes, have been used as novel and promising approaches. Furthermore, the present challenges and perspectives of using microbial hosts to produce ginsenosides have been discussed.

The parotoid gland secretion of toads of the family Bufonidae contain numerous pharmacologically active biocompounds. Rhaebo guttatus (R. guttatus) is a common species of this family in Brazil; however, this species has been poorly studied. Thus, the present study aimed to assess the immunomodulatory activity of a methanol extract from the parotoid gland secretion of R. guttatus in Swiss mice. Non-isogenic Swiss mice were administered (100 µl/mouse/day, orally) of the R. guttatus extract (8, 16, or 32 µg/ml), the vehicle (0.5% Tween-20, vehicle group), or distilled water (control group) for 7 or 30 days. After the treatment period, the mice were sacrificed, and the hearts, liver, lungs and kidneys were removed for histopathological analysis. Cells from peritoneal lavage and spleen were used for immunological analyses. The results revealed that the treatment did not affect water or feed consumption or body weight of the mice. The lungs of mice treated with 8 µg/ml for 30 days exhibited a reduction in relative weight compared to those of the control group. Histopathological analysis revealed that the extract induced edema, blood clots and leukocyte infiltrates, mainly after 30 days at doses of 8 and 32 µg/ml. The administration of the extract promoted the production of the pro-inflammatory cytokines, IL-12 and TNF-α, and reduced the production of IL-10 and IL-4. The spontaneous release of hydrogen peroxide by peritoneal macrophages was also induced in the mice treated with 16 µg/ml extract for 30 days. On the whole, the present study demonstrates that a crude methanolic extract from the parotoid gland secretion of R. guttatus has immunomodulatory activity and low toxicity, and may have potential as a stimulator of the immune system for immunosuppression conditions.
