Biological and phylogenetic characteristics of West African lineages of West Nile virus

Abstract: The West Nile virus (WNV), isolated in 1937, is an arbovirus (arthropod-borne virus) that infects thousands of people each year. Despite its burden on global health, little is known about the virus’ biological and evolutionary dynamics. As several lineages are endemic in West Africa, we obtained the complete polyprotein sequence from three isolates from the early 1990s, each representing a different lineage. We then investigated differences in growth behavior and pathogenicity for four distinct West African li… Show more

“…WNV L1 was first isolated in Africa [ 5 , 6 ] from where it spread to the Middle East and Europe during the mid-twentieth century, and finally to the United States (US) in 1999 [ 7 , 8 , 9 ]. Since 2004, a new WNV L2 is circulating in Europe [ 1 , 10 ] and is responsible for the unprecedented increase of WNV human cases registered since 2018 [ 11 , 12 , 13 ].…”

“…West Nile virus (WNV) is an enveloped positive-sense single stranded RNA virus of the genus Flavivirus, family Flaviviridae, and it is classified in the Japanese Encephalitis (JE) antigen complex, with Japanese Encephalitis Virus (JE) and Saint Louis Encephalitis Virus (SLEV) (Simmonds et al, 2017;Castillo-Olivares & Wood, 2004). Currently, the virus is classified into nine lineages (L1 -L9), with lineages 1 and 2 being the most described and associated with outbreaks of neurological disease in humans and horses (Fall et al, 2017).…”

“…Season 2018 has been very favorable for mosquitoes development; the dry period started with high temperatures unusually early in May (as opposed to 2019), extended into deep autumn and exceeded the total number of cases in all previous years (between 2010 and 2017) (Haussig et al, 2018). Important reason is that at least eight phylogenetic lineages of virus have been described (Fall et al, 2017), but only lineage 1 and 2 are associated with disease in humans (Petersen, 2013). In Europe, prior to the emergence of a lineage 2 in Hungary in 2004, sporadic cases and occasional outbreaks in animals and humans were due to lineage 1.…”
