Biological characteristics of maize dwarf mosaic potyvirus from Spain

Abstract: Two isolates of maize dwarf mosaic virus (MDMV-Sp and MDMV-Spl) obtained from maize in the northeast of Spain were studied. Both isolates induced similar reactions on 6 sorghum cultivars, johnsongrass and oat (cv. Clintland), with the exception of MDMV-Sp which produced a different reaction on one sorghum cultivar. Thirtythree grass species were tested as possible hosts (16 previously untested) and 18 were found to be susceptible. Among those, eight were previously unidentified hosts for MDMV: Aegilops ovata, … Show more

“…In this sense, it is remarkable that Sitobion avenae was the aphid species with the highest densities on Bt-maize compared to non-Bt-maize. This aphid transmits SCMV (42%) (Achon et al 2003) more efficiently than MDMV (6%) (Achon et al 1996). It will be necessary to study the relation between S. avenae populations and SCMV incidence over a longer period in order to confirm the positive effect of this aphid on the increasing presence of SCMV in Spain.…”

“…All samples were analyzed with polyclonal antisera against MDMV-Sp. (Achon et al 1996), SCMV (Loewe GmbH) and MRDV (Bio-Rad, Hercules, CA). Immunoglobulin (Ig)Gs from commercial antisera were used at the dilution and in the buffers recommended by the manufacturer, while in the same buffers, those isolated from antisera obtained in our laboratory were used at 0.1 lg/ml and alkaline phosphatase-conjugated IgGs were used at a dilution of 1:1,000.…”

“…To assess the putative bottleneck of aphid transmission in the MDMV population, haplotypes E and Y, which had both been obtained from Johnsongrass, were mechanically inoculated onto maize plants as described in Achon et al. (1996), and the resulting infected plants used to feed aphids (Table 8).…”

Transmission was carried out in a nonpersistent manner and transmission rates were determined serologically as indicated in Achon et al (2003).…”
