Biological Heterogeneity of Chondrosarcoma: From (Epi) Genetics through Stemness and Deregulated Signaling to Immunophenotype

Abstract: Chondrosarcoma (ChS) is a primary malignant bone tumor. Due to its heterogeneity in clinical outcomes and resistance to chemo- and radiotherapies, there is a need to develop new potential therapies and molecular targets of drugs. Many genes and pathways are involved in in ChS progression. The most frequently mutated genes are isocitrate dehydrogenase ½ (IDH1/2), collagen type II alpha 1 chain (COL2A1), and TP53. Besides the point mutations in ChS, chromosomal aberrations, such as 12q13 (MDM2) amplification, th… Show more

“…At least 460 distinct COL2A1 mutations, 663 distinct probands, and 21 illnesses have been identi ed so far 20 . One of the most often altered genes that contribute to the development of chondrosarcoma is COL2A1 21 . COL2A1 was a risk factor in our model as a result.…”

“…M1‐type macrophages demonstrate a proclivity for secreting inflammatory cytokines, thereby exerting anti‐tumour effects. In contrast, M2‐type macrophages, characterized by their secretion of anti‐tumour cytokines, paradoxically contribute to tumour progression 59 . Notably, within the TME, TAMs often exhibit a propensity for polarization towards the M2 type 59 .…”

“…Chondrosarcoma tumor tissue, like hyaline, is characterized by a dense ECM with poor blood and lymph vascularity, on which a low percentage of dividing cells is embedded. Thus, the ECM forms a physical semi-permeable barrier, inhibiting cytotoxic agents reaching their target, i.e., chondrosarcoma cells, while reduced blood circulation creates severe chronichypoxia [116]. Moreover, the Schwan chondrosarcoma ECM disturbance by modifying the synthesis of ECM components, mainly PGs, attenuates this tumor growth [117].…”
