Biological investigation of a supersaturated self-nanoemulsifying drug delivery system of Piper cubeba essential oil

Abstract: In this work, the mechanism of Piper cubeba essential oil anti-inflammatory activity alone and as a supersaturated self-nanoemulsifying drug delivery system (S-SNEDDS) was evaluated.

“…The results showed no significant changes in particle size, PDI, and % release of the optimized formulation F4, suggesting that the medicated optimized nanosuspension of MA was sufficiently stable. The stability of nanosuspensions has already been proved previously in the literature [28,[31][32][33][34][35]. Several drugs, such as cyadox, aprepitant, and azoxystrobin, were found to be stable when formulated into nanosuspension formulations [28,31,35].…”

“…The zeta potentials of the formulations F1-F8 were found to be −17 to −30 mV. The zeta potential value at a magnitude of ±30 mV suggested the maximum stability of nanoformulation [33]. Hence, the formulation F4 was found to be maximally stable compared with the other formulations studied.…”

“…Gastroprotective, analgesic, anti-inflammatory, and hepatoprotective activity of PO has been investigated in literature (Choi and Hwang 2003;Perazzo et al 2003;Morikawa et al 2004;AlSaid et al 2014AlSaid et al , 2015. More recently, mechanistic anti-inflammatory effects of PO at molecular level have also been reported in animal models (Shakeel et al 2015).…”

“…Nanomedicine-based drug delivery systems have great potential in enhancing pharmacological effects and pharmacokinetic profile of drug molecules and plant-based bioactive materials (Jaiswal et al 2015;Shakeel et al 2015;Odei-Addo et al 2017;Thiruvengadam et al 2018;Alam et al 2018). Recently, oral nanoemulsions and "self-nanoemulsifying drug delivery systems (SNEDDS)" have also been evaluated for the enhancement of pharmacological effects of several therapeutic agents (Jain et al 2015;Jaiswal et al 2015;Shakeel et al 2015;Sharma et al 2015;Cui et al 2019;Eleftheriadis et al 2019). SNEDDS are preconcentrates of nanoemulsions in which the drug is encapsulated in oil phase in the presence of surfactant/cosurfactant which could form very fine nanosized droplets/nanoemulsions upon mild agitation with an aqueous media (Villar et al 2012;Jain et al 2015;Shakeel et al 2015;Sharma et al 2015).…”

“…Hence, the objective of this research work was to develop suitable nanoemulsion formulations of EEO and to investigate its wound healing effects in rat model as compared with pure EEO and standard antibiotic gentamycin after oral administration. Macroemulsions were not prepared in this work because nanoemulsions have lower droplet size in comparison with macroemulsions and these systems are more suitable for enhancing absorption and therapeutic activity of essential oils 11,12,27 . Various nanoemulsion formulations of EEO were prepared using aqueous phase titration method via construction of pseudoternary phase diagrams.…”
