Biological Significance of Proliferation, Apoptosis, Cytokines, and Monocyte/Macrophage Cells in Bone Marrow Biopsies of 145 Patients With Myelodysplastic Syndrome

Abstract: Labeling index (LI), apoptosis, levels of 2 pro-apoptotic cytokines tumor necrosis factor-alpha (TNF-alpha) and transforming growth factor-beta(TGF-beta), and the number of monocyte/macrophage cells that are the likely source of the cytokines were simultaneously measured in plastic-embedded bone marrow (BM) biopsy sections of 145 patients with myelodysplastic syndromes (MDS). TNF-alpha was correlated with TGF-beta (P = .001) and with monocyte/macrophage cells (P = .003). Patients with excess blasts in their ma… Show more

“…25 Instead, it is possible that cytokines such as TGF-b, known to downregulate the surface expression of NKG2D and detected in high levels in patients with MDS, may be involved in the reduced expression of NKG2D. 39,40 Recently, increasing knowledge on the molecular specificities for NK-cell activation has been gained from experiments in reductionistic model systems. 18 Emerging evidence suggests that resting NK cells require signaling through multiple activating receptors simultaneously to induce natural cytotoxicity.…”

“…One such disorder is chronic myelomonocytic leukemia (CMML), originally categorized as MDS (18), but recently reclassified by the World Health Organization as a distinct entity (19). Indeed, ex vivo studies showed low TNF␣ levels in patients with CMML (20)(21)(22).…”

“…In the siltuximab group, the mean platelet count decreased from baseline (227. 6 ]; 21%). However, the incidence of thrombocytopenia was low and comparable between the two treatment groups (siltuximab group: 6%; placebo group: 4%).…”

“…In many patients with MDS, levels of pro-inflammatory cytokines, such as tumor necrosis factor-a (TNF-a), transforming growth factor-b (TGF-b), and interleukin-6 (IL-6), are elevated compared to healthy persons or patients with other forms of marrow failure [2][3][4][5][6][7]. In particular, IL-6 may play an important role in the pathophysiology of anemia of inflammation associated with MDS as co-stimulator of in vitro progenitor growth and mediator of disease progression in MDS [2,8,9].…”
