Biologics‐Loaded Photothermally Dissolvable Hyaluronic Acid Microneedle Patch for Psoriasis Treatment

Abstract: Emerging biologics, such as monoclonal antibodies (mAbs), are the most promising treatment for psoriasis, while their effective delivery to the local inflammatory area of the skin is a challenge. Here, a novel photo‐thermally dissolvable hyaluronic acid microneedle patch is presented with biologics IL‐17 mAbs and MXene encapsulation for psoriasis treatment. As the microneedles possess suitable mechanical strength and great biocompatibility, they can puncture the skin with minimal immune response. In addition, … Show more

“…Though MNs did not cover the whole ear, the released drugs can diffuse in the skins and transport to lymph nodes, thus inhibiting the progression of psoriasis in an area larger than patch size (Figure S21). 65,66 The mice in the MTX-HA group also had thinner ear and epidermal thicknesses than those of the untreated groups, indicating the released MTX had therapeutic effect for psoriasis. However, the treatment in MTX-HA and MTX/ EGCG-HA was less efficient than in the EGCG-HP and MTX/ EGCG-HP groups demonstrating the advantages of sustained and intelligent release of EGCG in psoriasis management.…”

“…Light is a frequently used exogenous stimulus in the clinical application of various diseases [102][103][104][105]. Due to the advantages of spatiotemporal selectivity, remote controllability, noninvasiveness, and convenient operation, lightresponsive DDSs based on phototherapies including photoactivated chemotherapy [106,107], photodynamic therapy (PDT) [108][109][110][111], photothermal therapy (PTT) [112][113][114], etc., have been broadly used for cancer treatment [115,116]. By using the light-controllable DDSs, on-demand drug/ gene/protein release could be achieved for precision strikes against cancer cells.…”

“…Transdermal drug delivery is an effective route for the treatment of skin diseases, because it allows direct targeting of lesions on the skin and reduces the side effects associated with systemic drug delivery (90)(91)(92)(93). However, the skin barrierstratum corneum could greatly impair the bioavailability of transdermal drug delivery.…”

“…unique advantages, microneedles have been widely used as effective transdermal drug delivery tools for the treatment of skin diseases including psoriasis (90,91,94). Du et al developed a microneedle patch made with hyaluronic acid (HA) and encapsulated MTX into the patch, which could effectively penetrate the IMQ-induced mouse epidermis and impair psoriasis-like skin inflammation in mice (95).…”
