Biomarkers in newly diagnosed pediatric-extensive chronic graft-versus-host disease: a report from the Children's Oncology Group

Abstract: Numerous chronic graft-versus-host disease (cGVHD) biomarkers have been identified in limited, single-institution studies without validation. We hypothesized that plasma-derived biomarkers could diagnose, classify, and evaluate response in children with cGVHD. We performed a concomitant analysis of a number of known and predicted peripheral blood cGVHD biomarkers from a Children's Oncology Group (

“…In this present study, there existed interindividual difference of cleaved BAFF levels (serum BAFF levels), and there was no statistical significance between different groups. And these results were different from observations on GVHD (17,18). Some factors in body fluid may affect the concentration and bioactivity of the proteolytic enzyme in kidney transplant recipients or stem cell transplant recipients.…”

“…Sarantopoulos et al concluded that elevated BAFF levels contribute to B cell activation in patients with active chronic GVHD after having tested whether BAFF correlated with chronic GVHD (cGVHD) disease activity and time of onset after allogeneic hematopoietic stem cell transplantation (17). Fujii et al performed a concomitant analysis of a number of known and predicted peripheral blood cGVHD biomarkers from a Children's Oncology Group (COG) phase III cGVHD therapeutic trial, and found that soluble BAFF (sBAFF) was one of four biomarkers with high specificity and sensitivity on onset of cGVHD (18).…”

“…As discussed being a cumbersome and time consuming technique 2DE could not reach the clinics, however, Mass spectrometry, especially MALDI-TOF have successfully made its way to pathology labs in the present decade. Several diseases like cancer, Alzheimer's disease, Amyloid lateral sclerosis, renal amyloidosis, and Severe combined immunodeficiency can now be diagnosed using mass spectrometry and its variants [28][29][30][31][32][33][34].…”

“…The cytokines were chosen as their levels have been reported to be associated with cGVHD and/or acute GVHD (aGVHD being a strong predictor of cGVHD). [21][22][23][24][25][26][27][28][29] g-glutamyl transpeptidase, cholinesterase, total protein and urea were chosen as their levels early post transplant had been reported to predict non-relapse mortality 20,30 and cGVHD is a major contributor to non-relapse mortality. ATG was chosen as its levels early post transplant have been reported to predict aGVHD and/or cGVHD.…”
