Biomarkers of oxidative stress in saliva in pigs: analytical validation and changes in lactation

Abstract: Background Biomarkers of oxidative stress in pigs have been measured in serum/plasma samples. However, blood collection in pigs can be highly stressful to the animals. Saliva is a biological fluid with several advantages in pigs over blood, since it can be easily collected without stress to the animals, being therefore an ideal sample in this species. The objective of this study was the validation of assays for the evaluation of oxidative stress status in saliva of pigs. For this purpose, three as… Show more

“…The ELISA used for quantitative determination of adrenaline (epinephrine) (Adrenaline Research ELISA™) had intra-assay and inter-assay variability less than 10% and a limit of detection of 0.25 ng/ml. Total oxidant status, paraoxonase 1 (PON1) and total antioxidant capacity (TAC; measured by trolox equivalent antioxidant capacity) were analysed following published methods [27][28][29]. All determinations of oxidative stress biomarkers were performed in an automated biochemistry analyser (Olympus AU600 Automatic Chemistry Analyzer, Olympus Europe GmbH, Germany).…”

“…Although serum is usually used to measure biomarkers that can provide information about oxidative status in pigs [ 44 , 45 ], saliva can also be used in this species [ 46 ] with the advantage of being a sample that can be easily collected without producing stress to the pigs.…”

“…These biomarkers were measured in both serum and saliva in order to assess the possible use of saliva as an alternative or complement to serum in oxidative status evaluation. In saliva, the same biomarkers as serum were measured except for thiol and TOS because, to the author’s knowledge, there are currently no methods that can accurately measure these biomarkers in pig saliva [ 46 ].…”

“…To measure the antioxidant levels in serum and saliva, three different spectrophotometric methods were used: cupric reducing antioxidant capacity (CUPRAC); ferric reducing ability of plasma (FRAP); Trolox equivalent antioxidant capacity (TEACH) [34]. In all cases, the resulting data were expressed as millimoles of Trolox equivalents per liter (mmol/L).…”
