SummaryThe high storage capacities and excellent biocompatibilities of metal-organic frameworks (MOFs) have made them emerging candidates as drug-delivery vectors. Incorporation of surface functionality is a route to enhanced properties, and here we report on a surface-modification procedure—click modulation—that controls their size and surface chemistry. The zirconium terephthalate MOF UiO-66 is (1) synthesized as ∼200 nm nanoparticles coated with functionalized modulators, (2) loaded with cargo, and (3) covalently surface modified with poly(ethylene glycol) (PEG) chains through mild bioconjugate reactions. At pH 7.4, the PEG chains endow the MOF with enhanced stability toward phosphates and overcome the “burst release” phenomenon by blocking interaction with the exterior of the nanoparticles, whereas at pH 5.5, stimuli-responsive drug release is achieved. The mode of cellular internalization is also tuned by nanoparticle surface chemistry, such that PEGylated UiO-66 potentially escapes lysosomal degradation through enhanced caveolae-mediated uptake. This makes it a highly promising vector, as demonstrated for dichloroacetic-acid-loaded materials, which exhibit enhanced cytotoxicity. The versatility of the click modulation protocol will allow a wide range of MOFs to be easily surface functionalized for a number of applications.

Docetaxel (Doc) is an antitumor drug used to treat various types of cancers. However, it has certain limitations in clinical use due to its side effects. The major objective of this study was to assess the antitumor activity and cardiotoxicity of the Doc loaded in a nanoemulsion formulated with the garlic oil (Doc-NEGO) in Ehrlich ascites carcinoma (EAC)-bearing mice. One hundred mice were split into five groups (n = 20). Groups I and II served as untreated mice and untreated EAC-bearing mice, respectively. Groups III-V represented the EAC-bearing mice administered orally by Doc-NEGO, NEGO and Doc dissolved in distilled water, respectively. The antitumor activity included the measurement of the mean survival time (MST) and the lactate dehydrogenase (LDH) activity in the tumor ascetic fluid. The cardiotoxicity was identified by determining the serum enzymes and analyzing the oxidative stress in the heart tissue. It has been found that the z-average diameters of the nanoparticles of NEGO and Doc-NEGO, determined by the zetasizer, were 63.19 ± 1.85 nm and 110.43 ± 14.37 nm, respectively. The administration of NEGO in the EAC-bearing mice has enhanced the LDH activity in the ascetic fluid and ameliorated the heart enzymes when compared to the other groups. Treating the EAC-bearing mice with the Doc-NEGO has improved the MST of the mice (27.7 ± 11.63 days) relative to the MST of EAC-bearing mice treated with Doc-water (23.1 ± 1.52 days). In conclusion, NEGO holds a great potential as a nanocarrier for the Doc in improving its efficacy and eliminating its cardiotoxicity.

Combined therapy is a promising strategy for clinical cancer treatment with synergistic effects. The purpose of the work reported was to evaluate a smart nanocarrier for co‐delivery of doxorubicin (DOX) and methotrexate (MTX). Since star‐like nanocarriers can load a high dose of drugs with various properties, we developed star polymer nanomicelles based on poly[(2,2‐dimethylaminoethyl methacrylate)‐block‐(2‐hydroxyethyl methacrylate)‐block‐(acrylic acid)] having potential for multi‐drug delivery. The nanomicelles demonstrated high encapsulation efficiency, i.e. 97.1% for DOX and 79.5% for MTX. To this end, the star‐like terpolymers were synthesized via atom transfer radical polymerization with pentaerythritol as an initiator. The micellar properties and dual stimuli‐responsive behaviour of the terpolymers were investigated using transmission electron microscopy, field emission scanning electron microscopy and dynamic light scattering measurements, concluding that this co‐therapy offers a promising approach for cancer treatment. © 2019 Society of Chemical Industry
