Biomimetic CO2Sequestration: 1. Immobilization of Carbonic Anhydrase within Polyurethane Foam

Abstract: Bovine carbonic anhydrase (CA) was immobilized within polyurethane (PU) foam for biomimetic CO 2 sequestration. The catalytic activities for the free and immobilized CA were estimated using paranitrophenyl acetate (p-NPA) as the substrate. Because the p-NPA has limited solubility in the aqueous phase, the activities were estimated in Tris buffer containing 10% acetonitrile. A Lineweaver-Burk relationship was employed to estimate the Michaelis-Menten kinetic parameters for the free and immobilized CA. The k cat… Show more

“…Numerous studies have investigated immobilizing CA onto a solid medium in order to increase its lifespan, particularly by minimizing exposure to denaturing conditions that may exist in an industrial carbon sequestration scenario (Cheng et al, 2008;Ozdemir, 2009;Sharma et al, 2011;Vinoba et al, 2012Vinoba et al, , 2011aWanjari et al, 2012;Yadav et al, 2011Yadav et al, , 2010. With particular relevance to this study, BCA immobilized within polyurethane foam preserved 100% activity over 45 days when stored in Tris buffer at ambient conditions in comparison to free BCA that lost its activity over the same period at 4 • C (Ozdemir, 2009).…”

“…The high porosity of PUF can reduce the diffusion resistance and provide good mass transfer. Moreover, elasticity and flexibility can provide stability and resistance to the compression set, which are interesting properties for industrial application [9][10][11].…”

“…S2) is a kind of inert material with excellent mechanical properties, such as high resistance and elasticity, bulk availability, and very low commercial cost [19,23]. PUF has high porosity (near 97 %) and a large adsorption surface.…”
