Biomimetic Membrane System Composed of a Composite Interpenetrating Hydrogel Film and a Lipid Bilayer

Abstract: International audienceA simple way to form an interpenetrating hydrogel (IPH) by combining a layer-by-layer polyelectrolyte membrane with agarose is reported. The formed IPH membrane is more robust and easily manipulated compared to a polyelectrolyte membrane with the same number of layers. The IPH has good diffusion characteristics and retains the advantageous surface charge from the polyectrolyte composition that facilitates the adsorption of a stable lipid bilayer. The stable adsorption of a lipid bilayer o… Show more

“…The IPH was derived from the combination of a layer-by-layer polyelectrolyte membrane with agarose. The resulting IPH is about 300 nm, thicker and rougher compared to a simple layer-by-layer polyelectrolyte membrane but the IPH retains the surface charges, which assist the adsorption of a stable lipid bilayer [20] . With only VDAC inserted into the system, the advantage of the immuno-SEM preparation is the absence of cross-reactions with other proteins.…”

“…The PEM sample was prepared using the layer by layer (LbL) technique to deposit PSS-PAH on agarose as described in Stidder et al [20] and transferred to a hydrophobic To avoid the "charging effect" when sample was analysed at high resolution, the membrane was coated with a thin layer of platinum. In addition, some chemical analysis was performed by X ray spectrometry (EDS) using a Bruker SDD detector.…”

“…A tethered lipid bilayer membrane (tBLM) is a planar membrane composed of mobile lipids and molecules tethered to a solid planar substrate that provides a realistic model of a biomembrane. The tBLM is stable due to the chemical attachment of the tethering molecules to the substrate . For these experiments the tethering molecules were those described previously as the molecules of benzyl disulfide undecaethylene glycol phytanol (DLP) and benzyl disulfide tetraethylene glycol polar spacer molecules dissolved in ethanol at a concentration of 400 μM and mixed in the ratio of 10:90.…”

“…The tBLM is tethered to the gold electrode through the covalent attachment of hydrophobic spacer molecules with a hydrophilic linker. The tBLM is coupled to a potentiostat for electrical impedance spectroscopy (EIS) measurements . This device is usually used to measure the conductive properties of membrane transport proteins .…”
