Assessment of in vitro antidiabetic properties of synthesized silver nanoparticles using ethanolic extract of Boerhavia diffusa

The in vitro antidiabetic efficacy of ethanolic extract Boerhavia diffusa (B.diffusa) synthesized silver nanoparticles (AgNPs) was investigated by inhibition of α-amylase, α-glucosidase, protein glycation assay, non-enzymatic glycosylation of hemoglobin, glucose uptake by yeast cells and glucose diffusion at varying concentrations (10 to 100µg/ml). The alpha-amylase assay shows that the acarbose (standard) and B. diffusa had IC50 values of 46.2 µg/ml and 55.4 µg/ml, whereas alpha-glucosidase inhibitory activity was found to be 63.4 µg/ml and 93.0 µg/ml respectively. Further, non-enzymatic glycosylation analysis showed IC50 value of metformin (standard) as 28.6 µg/ml and B. diffusa as 63.9 µg/ml. The protein glycation activity was inhibited in non-enzymatic glycosylation of hemoglobin. The glycosylation was induced using pioglitazone (standard) which gives IC50 value of 616.4 µg/ml by which B. diffusa showed 756.3 µg/ml. The uptake of glucose by yeast cells was analyzed and the result shows that the glucose concentration increased steadily from 5mM to 25mM (maximum absorption) of both metronidazole (standard) and B. diffusa. From 30 to 180 minutes, the glucose diffusion experiment revealed that the concentration of the metformin and B. diffusa extract was positively correlated with the time. The ethanolic extract of synthesized AgNPs and the reference medication employed in all experiments both benefit their curative potential for the treatment of insulin resistance. The generated silver nanoparticles can be used for industrial and therapeutic purposes and can be released into the environment without harm. More in vivo study can be reviewed, however the green synthesized ethanolic extract of B. diffusa exhibits promising affect for the treatment of diabetes mellitus.

One of the tomato’s acutely devastating diseases is Alternaria leaf spot, lowering worldwide tomato production. In this study, one fungal isolate was isolated from tomatoes and was assigned to Alternaria alternata TAA-05 upon morphological and molecular analysis of the ITS region and 18SrRNA, endoPG, Alt a1, and gapdh genes. Also, Urtica dioica and Dodonaea viscosa methanol leaf extracts (MLEs) were utilized as antifungal agents in vitro and compared to Ridomil, a reference chemical fungicide. The in vitro antifungal activity results revealed that Ridomil (2000 µg/mL) showed the highest fungal growth inhibition (FGI) against A. alternata (96.29%). Moderate activity was found against A. alternata by D. viscosa and U. dioica MLEs (2000 µg/mL), with an FGI value of 56.67 and 54.81%, respectively. The abundance of flavonoid and phenolic components were identified by HPLC analysis in the two plant extracts. The flavonoid compounds, including hesperidin, quercetin, and rutin were identified using HPLC in D. viscosa MLE with concentrations of 11.56, 10.04, and 5.14 µg/mL of extract and in U. dioica MLE with concentrations of 12.45, 9.21, and 5.23 µg/mL, respectively. α-Tocopherol and syringic acid, were also identified in D. viscosa MLE with concentrations of 26.13 and 13.69 µg/mL, and in U. dioica MLE, with values of 21.12 and 18.33 µg/mL, respectively. Finally, the bioactivity of plant extracts suggests that they play a crucial role as antifungal agents against A. alternata. Some phenolic chemicals, including coumaric acid, caffeic acid, ferulic acid, and α-tocopherol, have shown that they may be utilized as environmentally friendly fungicidal compounds.

Objective: This study aimed to assess the anti-rheumatic potential of Dodonaea viscosa and to evaluate its bioactive small molecules for their beneficial effects in the management of rheumatoid arthritis.Methods:In vitro bioactivity assays were performed to assess the healing potential of D. viscosa and statistical analysis was performed by using the linear regression technique. In silico analysis was performed to identify the key inhibitors of the disease to target TNF-α. The plant extract was prepared using ethanol solvent via the Soxhlet method. Phytochemical and bioactivity testing was performed. Gas chromatography–mass spectrometry (GC-MS) analysis was conducted for bioactive plant compounds. Disease-specific target was shortlisted by HUB gene analysis. Molecular docking and molecular dynamic simulations were run for validation of the results.Results: Phytochemical studies verified the presence of phenols, flavonoids, steroids, sterols, saponins, coumarins, tannins, and terpenoids. The significant antioxidant potential of plant extract was evaluated by the DPPH and Ferric Reducing Antioxidant Power (FRAP) assays, while the anti-inflammatory potential was evaluated by the protein denaturation and Human Red Blood Cell (HRBC) membrane stabilization assays. In silico studies revealed that nine of the 480 compounds found in D. viscosa (ethanol extract) had drug-like properties. Tumor necrosis factor alpha (TNF-α) was selected as a key disease gene through HUB gene analysis. Results of molecular docking and MD simulation analysis demonstrated that 4-(1-hydroxy-3-oxo-1H-isoindol-2-yl) benzoic acid (PubChemID 18873897), had the best binding affinity with TNF-α amongst all nine compounds.Conclusion: 4-(1-hydroxy-3-oxo-1H-isoindol-2-yl) benzoic acid (PubChemID 18873897), have the potential to be a good small molecule inhibitor of TNF-α against rheumatoid arthritis.
