Objectives: The main objective of this study was to evaluate the activity of enzymes with hydrocarbon-degrading potential. Methods: Four petroleum hydrocarbon-degrading bacterial strains, S4P2, S14P1, S9D2, and S13D1, were isolated and used to screen the activity of catalase, lipases, and catechol 1, 2-dioxygenase. All the strains were characterized biochemically. The presence of catalase was studied using nutrient agar and absorbance was measured at 240 nm due to the disappearance of peroxide. Qualitatively, the presence of lipase was studied on tributyrin agar (TBA) plates. The lipase activity was quantified using the titrimetric method. The activity of catechol 1, 2-dioxygenase (C12O) was screened by the formation of the product cis, cis-muconic acid (CCMA) by measuring the absorbance at 260 nm. Findings: Bacterial strains S4P2, S14P1, S9D2, and S13D1 showed clear halo zones around TBA plates, indicating lipase activity. Similarly, quantitively lipase activity was highest in S4P2 (0.034±0.01 U/ml) and S13D1 (0.027±0.01 U/ml) followed by S9D2 (0.024±0.01 U/ml) and S14P1 (0.024±0.01 U/ml) respectively. The presence of catalase was confirmed by effervescence after the addition of H2O2 on cultured nutrient agar, quantitative screening was performed by checking the decrease in absorbance at 240 nm. The maximum activity was observed in isolates S9D2 and S14P1. The total catechol 1, 2-dioxygenase activity was found to be 18.48±0 U/ml and 8.96±0.56 U/ml in S9D2 and S13D1. In contrast, S14P1 and S4P2 showed 12.88±0.56 U/ml and 16. 8±0.U/ml. Novelty: Microbial enzymes play an important role in the degradation of hydrocarbons. The direct application of microbes offers many constraints, so the use of these enzymes as a cocktail may be a solution for bioremediation. In the present study, we aimed to elucidate the enzymes responsible for hydrocarbon degradation in previously unreported strains, Pseudomonas donghuensis and Azospirillum zeae. We have earlier isolated Aspergillus luchuensis, A. violaceofuscus, A. niveus, A. terreus, A. japonicus and A. ustus from naturally degraded diesel oil in the soils using nutrient rich technique. These species were cultured to produce enzyme complexes on 10% (w/v) diesel oil broth containing 0.5 and 1.0% (w/v) jaggery at 37 and 45℃ temperature separately for 7, 14 and 21 days. It was observed that all 6 species of Aspergillus produced luxuriant mycelial growth at day-7 on both temperatures, however, at 45℃, the liquid broth dried on 14th day of incubation, and therefore, secretory extracellular enzymes produced at 37℃ were screened to test the activities of amylases, gelatinases, lipases, cellulases, pectinases, proteases and manganese peroxidases using agar plate assay method. These enzymes are responsible for beaking down different bonds of the complex hydrocarbons. It was found that all 6 test species of Aspergillus produced all 7 enzymes at higher concentration at day 21 on diesel oil containing 1.0% jaggery. The production of enzyme increased with increasing period of incubation from day 7 to day 21. A. luchuensis produced highest activities of proteases, A. violacefuscus - amylases, A. niveus - gelatinases, A. terreus and A. ustus - pectinases, and A. japonicus produced highest activities of manganese peroxidases. All these enzymes contribute in the degradation of diesel oil in nature. This study revealed that the production of varying amounts of different enzymes by all these 6 species of Aspergillus has significant ecological and bioremediation potential.

The high global bacterial infection burden has created need to investigate the neglected potential drivers of pathogenic bacteria, to inform disease prevention. Kampala is facing a proliferation of herbalists, selling herbal medicine (HM), of largely unregulated microbiological quality. We evaluated the bacterial contamination burden in HM sold in Kampala, to support evidence-based redress. The total viable loads (TVL), total coliform counts (TCC), E. coli counts, and prevalence of selected bacterial strains in 140 HM were examined using conventional culture, following the guidelines of World Health Organization (WHO), and Uganda National Drug Authority (NDA). Data were analyzed using D'Agostino-Pearson test, frequencies, proportions, Chi-square, and Mann–Whitney U test with STATA version-15.0. Fifty (35.7%), fifty-nine (42.1%), and twelve (8.6%) HM were unsafe for human use because they exceeded WHO’s permissible limits for TVL, TCC, and E. coli counts respectively. Solids had significantly higher mean TVL than liquids. Violation of NDA’s guidelines was significantly associated with high TVL. Fifty-nine bacteria, viz., Klebsiella pneumoniae (n = 34; 57.6%), Escherichia. coli (12; 20.3%), Staphylococcus aureus (7; 11.9%), Klebsiella oxytoca (3; 5.1%), Bacillus cereus, Pseudomonas aeruginosa, and Enterobacter spp. (1; 1.7% each), were isolated from 45 (32.1%) samples. These bacteria can cause severe clinical diseases, and promote deterioration of HM potency.
