Biosynthesis and Secretion of Serine Peptidase SerP38 from Tenebrio molitor in the Yeast Komagataella kurtzmanii

Abstract: A strain of the yeast Komagataella kurtzmanii, a producer of recombinant peptidase SerP38 from the yellow mealworm Tenebrio molitor, has been obtained. The level of proenzyme secretion was 20-50 mg/L. It was shown that the target His 6 -tagged protein was produced in two forms during secretion in yeast. One of them was a monomer that was efficiently purified via Ni-NTA chromatography and then activated with trypsin. Another form accumulated in the culture medium as oligomers prone to aggregation in the presenc… Show more

“…Most of them are in the "S" group, many of which act in digestion (Bao et al, 2014;Cao & Jiang, 2018;Prabhakar et al, 2007;Wolfson et al, 1990). Additionally, they may possess a prominent role in rapid dietary adaptations (Gorbunov et al, 2021;Kumar et al, 2021). Similarly, transcripts of 14 SPs/SPHs are most abundant in salivary glands, where they may also act in digestion (Allen & Walker, 2012;Amino et al, 2001;Walker et al, 2018).…”

“…Analyzing trypsin-like SPs/SPHs in transcriptome datasets from different stages of the T. molitor life cycle, we have previously de novo assembled 54 trypsins and five trypsin-like SPHs [ 31 ]. We also characterized recombinant preparations of SP, SerP38, and SPH, SerPH122, expressed in the Komagataella kurtzmanii system [ 32 , 33 ]. Recent work by Wu and coauthors [ 34 ] provided information on 200 T. molitor genes including 112 SPs and 88 SPHs, and transcriptome datasets together with RT-PCR analysis were used for SP-related genes expression profiling at various developmental stages and tissues.…”

“…This strain was also registered in CBS under the number CBS 12817 and included in the collection of Komagataella species selected for genotypic and phenotypic diversity analysis (Heistinger et al, 2022). A competitive recombinant expression system was developed and successfully applied based on K. kurtzmanii VKPM Y‐727 (Gorbunov et al, 2021; Gubaidullin et al, 2019).…”
