Biosynthesis of Panaxynol and Panaxydol in Panax ginseng

Abstract: Abstract:The natural formation of the bioactive C 17 -polyacetylenes (−)-(R)-panaxynol and panaxydol was analyzed by 13 C-labeling experiments. For this purpose, plants of Panax ginseng were supplied with 13 CO 2 under field conditions or, alternatively, sterile root cultures of P. ginseng were supplemented with [U- 13C 6 ]glucose. The polyynes were isolated from the labeled roots or hairy root cultures, respectively, and analyzed by quantitative NMR spectroscopy. The same mixtures of eight doubly 13 C-labeled… Show more

“…1). Early intermediates of this pathway have been identified by radiochemical studies (Barley et al, 1988;Knispel et al, 2013), and the pathway has been examined in diverse campanulid lineages (Lee et al, 1998;Cahoon et al, 2003;Carlsson et al, 2004;Nam and Kappock, 2007;Busta et al, 2018). Linoleic acid is converted to crepenynic acid by installing the first acetylenic bond in the D 12 position (the 12 th carbon relative to the fatty acid head group; this notation is used consistently throughout).…”

“…In close similarity to the labeling pattern of the fatty acid residue of the lupeol derivative, a strict polyketide pattern was also observed for 13 CO 2 -labeled panaxynol and panaxydol from the plethora of secondary products formed by Panax ginseng [ 55 ]. Specifically, the number of 13 C atoms that had been imported jointly does not exceed two in any case.…”

“…The use of 13 CO 2 as a tracer is a powerful tool, since this strategy represents the possibility of studying metabolic pathways in plants by mimicking, as much as possible, their physiological growth conditions [ 40 , 41 , 42 ]. The labelling period (pulse) is followed by a chase period in which the plants grow under normal conditions and natural 13 CO 2 abundance for several hours/days.…”
