Blockage of Mouse Muscle Nicotinic Receptors by Serotonergic Compounds

Abstract: summaryXenopus laevis oocytes were used to analyse the effects of serotonin (5-hydroxytryptamine, 5_HT) and serotonergic agents on ionic currents elicited by the activation of mammalian muscle nicotinic acetylcholine receptors (AChRs). 5_HT as well as other serotonergic agents, such as ketanserin, 8-hydroxy-2-(di-n-propylamino)tetralin (8-OH-DPAT), methysergide, spiperone, or fluoxetine alone (up to 1 mÒ), did not elicit membrane currents in Xenopus oocytes expressing AChRs, but they reversibly reduced the cur… Show more

“…The rank order of potency for inhibition (IC 50 ) by tryptamine homologues was 5‐methyltryptamine (12 μ M )>1‐methyltryptamine (111 μ M )>(±)5‐F‐ α ‐methyltryptamine (123 μ M )>2‐methyl‐5‐OH‐tryptamine (150 μ M )>6‐methyltryptamine (218 μ M )>7‐methyltryptamine (650 μ M ), suggesting that the group at the 5‐position may be an important structural motif that governs the potency of these compounds (Berger, 2000). 5‐HT has also been reported to modulate other ligand‐gated ion channels, including nicotinic ACh, ATP and GABA A receptors in vivo (Kawahara et al ., 1994; Koizumi et al ., 1995; Garcia‐Colunga & Miledi, 1999; Fucile et al ., 2002).…”

“…There is evidence that serotonin and serotonergic receptor acting drugs such as ketanserin can act directly on nicotinic receptors. Ketanserin and other serotonergic ligands as well as serotonin itself have been found to block and enhance desensitization of the muscle nicotinic receptor (Garcia-Colunga and Miledi, 1999). It would be interesting to see if this direct action of ketanserin occurs on neuronal nicotinic receptors as well.…”
