Blood Donors with Indeterminate Anti-p24^gag Reactivity in HIV-1 Western Blot: Absence of Infectivity to Transfused Patients and in Virus Culture

Abstract: During a follow-up period of 23-40 months, 7 regular blood donors had persistently, and 4 had intermittently indeterminate anti-p24^gag reactivity in human immunodeficiency virus (HIV)-l Western Blot. Serological testing and viral cultures revealed that these donors had no signs of infection for HIV-1, HIV-2, human T-cell lymphotropic virus (HTLV)-4, and HTLV-1. Extensive interviewing and physical examination of these donors revealed neither risk factors, nor signs of HIV infection in the tested donors. Ten re… Show more

“…Direct testing for HIV infectivity was carried out by cocultivation of donor-derived lymphocytes with permissive cells. The sum of these data shows that HIV-1 detection was restricted to Western blot analysis only [6,7].…”

“…No positive signal was found for any of these donors, either using EtBr staining, blotting and screening or using the LH assay. The complete description of these donors is given by van der Poel et al [6]. Figure 5 shows the negative results of one DNA sample of each of these donors after nested gag PCR followed by LH detection.…”
