Bloom Helicase and DNA Topoisomerase IIIα Are Involved in the Dissolution of Sister Chromatids

Abstract: Bloom's syndrome (BS) is an autosomal disorder characterized by predisposition to a wide variety of cancers. The gene product whose mutation leads to BS is the RecQ family helicase BLM, which forms a complex with DNA topoisomerase III␣ (Top3␣). However, the physiological relevance of the interaction between BLM and Top3␣ within the cell remains unclear. We show here that Top3␣ depletion causes accumulation of cells in G 2 phase, enlargement of nuclei, and chromosome gaps and breaks that occur at the same posit… Show more

“…Despite possessing seemingly identical relaxation activity, Top3␤ cannot substitute for Top3␣ in vivo (20,23) or in dHJ dissolution (2). The main structural difference between the two isoenzymes lies in their C termini.…”

“…Cells derived from BS patients show genomic instability, including the hallmark feature of an elevated frequency of crossover recombinationas scored by measuring SCE (Chaganti et al 1974;German 1993 (Li and Wang 1998;Plank et al 2005;Hartung et al 2007). In chicken DT40 cells, TopoIIIa depletion also causes cell death associated with dramatic genomic instability (Seki et al 2006). In Schizosaccharomyces pombe, deletion of top3 þ is also lethal and cell death is associated with chromosome missegregation (Goodwin et al 1999;Maftahi et al 1999;Oh et al 2002).…”

“…There are three pathways for HJ processing (5). In humans, the primary pathway involves the BLM helicase-TopoisomeraseIIIα-RMI1-RMI2 (BTR) complex, which promotes the dissolution of double Holliday junctions (dHJs) by catalyzing the convergent migration of each junction followed by topoisomerasemediated processing of the resulting hemicatenane (6)(7)(8). This pathway favors the formation of noncross-over products (NCOs).…”
