Body composition analysis of obesity and hepatic steatosis in mice by relaxation compensated fat fraction (RCFF) MRI

Abstract: Purpose To develop and validate a quantitative MRI methodology for phenotyping animal models of obesity and fatty liver disease on 7T small animal MRI scanners. Materials and Methods A new MRI acquisition and image analysis technique, Relaxation-Compensated Fat Fraction (RCFF), was developed validated by both Magnetic Resonance Spectroscopy and histology. This new RCFF technique was then used to assess lipid biodistribution in two groups of mice on either a high fat (HFD) or low fat (LFD) diet. Results RCF… Show more

“…5C(a)). Staining with PCNA demonstrated increased proliferative index of the glandular epithelium of the B6.V-Lepob/J mice, compared to C57BL/6N mice (23.26 AE 7.43 vs. 19.58 AE 5.52; P < 0.05], which might also contribute in increased prostate size ( Fig. 5C(b)).…”

“…A 72-mm diameter volume coil was used for excitation and signal detection to maximize the uniformity of the images. After localizer scans, a Relaxation Compensated Fat Fraction (RCFF) MRI acquisition and reconstruction process was used to generate quantitative fat fraction maps for each imaging slice [29]. Briefly, three asymmetric echo spin echo MRI acquisitions were acquired (TR/TE = 1500 ms/20 ms, 17–25 coronal slices, resolution = 200 um×200 um×1000 um, 2 averages).…”

“…A similar analysis was performed with a relaxation‐compensated fat fraction (RCFF) , which was calculated as previously described, and as follows: RCFF=true|normalρfattrue|true(1eTR/T1fattrue)eTR/T2fattrue|normalρwatertrue|true(1eTR/T1watertrue)eTR/T2fat+true|normalρfattrue|true(1eTR/T1fattrue)eTR/T2fat …”

“…The T 1 and T 2 values that were used in the description of RCFF were used here. The nominal TR and TE values, as delineated above, were also used in the calculation.…”
