Bone Marrow NK Cells: Origin, Distinctive Features, and Requirements for Tissue Localization

Abstract: NK cell maturation is a continuous process, which initiates in the bone marrow and proceeds in peripheral tissues, where NK cells follow distinct differentiation routes. Drastic phenotypic changes are observed during progression from precursors to mature NK cells, including changes of expression and functionalities of several chemoattractant receptors. Upon differentiation, mature NK cells migrate outside the bone marrow; as well, peculiar subsets of NK cells can also home back to or localize in this anatomic … Show more

“…These subsets include ILCs1, ILCs2, ILCs3, lymphoid tissue-inducer (LTi) cells, and conventional NK (cNK) cells [ 138 , 139 ]. ILCs interact with the tissue cells to ensure, in addition to the immune response, homeostasis and tissue repair [ 140 , 141 ]. Among these subsets, cNKs are very similar to Tbet + ILCs1 producing IFN-γ but differ in their higher cytotoxic potential.…”

“…Development and maturation of NK cells: NK cell maturation is a continuous process that is initiated in the BM and is continued in peripheral tissues [30]. Mature NK cells reside in the BM and secondary lymphoid tissues such as lymph nodes [7].…”

“…Mature NK cells reside in the BM and secondary lymphoid tissues such as lymph nodes [7]. Thus, BM is not only the place for development, maturation, self-renewal and persistence of NK cells, but also it provides a line of defense against infections and tumors through utilizing the presence of NK cells [30]. The stages of development of NK cells include: NK progenitor cells, immature NK cells, and mature NK cells.…”

“…One group of immune cells assumed to develop within the BM are natural killer (NK) cells [4], whereby NK cell development is also taking place at extramedullary sites [5]. NK cells are of great importance for the innate immune response as they are able to quickly and efficiently eliminate cells that have down-regulated human leukocyte antigen (HLA)-class I molecules on the cell surface, such as in damaged, malignant, and virus infected cells [6].…”
