Bovine CD2-/NKp46+ cells are fully functional natural killer cells with a high activation status

Abstract: Background: Natural killer (NK) cells in the cow have been elusive due to the lack of specific NK cell markers, and various criteria including a CD3 -/CD2 + phenotype have been used to identify such cells. The recent characterization of the NK-specific NKp46 receptor has allowed a more precise definition of bovine NK cells. NK cells are known as a heterogeneous cell group, and we here report the first functional study of bovine NK cell subsets, based on the expression of CD2.

“…2c). In accordance with this finding, bovine CD2 − NK cells have been shown to be preferentially activated when stimulated with interleukin‐2 31. Furthermore, the expression of a second activation marker, CD44, was found to be significantly increased in AL NK cells, which highlights an overall activated phenotype within AL 23.…”

“…The CD2 ϩ subset usually comprises a smaller proportion of the cells than the CD2 Ϫ subset in young animals (9). The role of NK and ␥␦ T lymphocytes in immunity and immunopathogenesis of JD is unclear, but they may be a source of cytokines that modulate immune responses (6,25,30,47). The majority of freshly isolated bovine NK cells express CD2.…”

“…Upon activation by the cytokines TNFα, IFNα/IFNβ, IL-12, IL-15 or IL-18 produced by accessory cell populations such as antigen presenting cells (APC), NK cells have two main effector functions. Firstly, NK cells are a significant source of immunoregulatory cytokines, primarily IFNγ but also TNFα, GM-CSF [ 97 99 ], IL-10 [ 98 , 99 ] and IL-22 [ 100 , 101 ]. Secondly, NK cells display cytotoxicity to target cells through the release of preformed, cytotoxic granules containing perforin and granulysin [ 102 ].…”
