Bovine Lactadherin as a Calcium-independent Imaging Agent of Phosphatidylserine Expressed on the Surface of Apoptotic HeLa Cells

Abstract: S U M M A R Y Bovine lactadherin holds a stereo-specific affinity for phosphatidylserine (PS) membrane domains and binds at PS concentrations lower than the benchmark PS probe, annexin V. Accordingly, lactadherin has recognized PS exposure on preapoptotic immortalized leukemia cells at an earlier time point than has annexin V. In the present study, the cervical cancer cell line HeLa has been employed as a model system to compare the topographic distribution of PS with the two PS binding proteins as adherent ce… Show more

“…PS exposure is a universal hallmark for cells undergoing apoptosis [17]. For this study, we used lactadherin to assess PS as lactadherin binds PS in apoptotic cells at an earlier time point than annexin V, the benchmark probe to assess PS exposure [14]. To differentiate between early and late apoptosis, we stained cells with propidium iodide (PI) [16].…”

“…Cell stress quantified by PS exposure was monitored over time using FITC labeled Lactadherin. Labeling was done as described previously [44], [49] with FITC-lactadherin and PI used as apoptosis marker in a similar fashion as Vermes et al, Fadok et al a.o. [59][61].…”

“…Lactadherin also displays stereospecific binding to phosphatidyl-L-serine and preference for convex membranes. Membrane binding of lactadherin is not Ca 2+ -dependent which further improves its value for monitoring and blocking exofacial PS [41], [44][46]. There are structural and functional similarities between the C1 and C2 domains of lactadherin and those of blood coagulation factors V and VIII [39], [41], [42].…”

“…In vitro , FITC-labelled bovine lactadherin has been used for early detection of apoptosis in leukaemia cell lines treated by etoposide, and also in HeLa cervical cancer lines treated by staurosporine [54,55]. HYNIC-lactadherin has also been successfully labelled with 99m Tc (i.e.…”
