Bovine pericardial extracellular matrix niche modulates human aortic endothelial cell phenotype and function

Abstract: Xenogeneic biomaterials contain biologically relevant extracellular matrix (ECM) composition and organization, making them potentially ideal surgical grafts and tissue engineering scaffolds. Defining the effect of ECM niche (e.g., basement membrane vs. non-basement membrane) on repopulating cell phenotype and function has important implications for use of xenogeneic biomaterials, particularly in vascular applications. We aim to understand how serous (i.e., basement membrane) versus fibrous (i.e., non-basement … Show more

“…To answer this question, endothelial cells were seeded on the fibrous layer of the GA-treated pericardium, which in the native valve is the most proximal layer contacting blood [29]. Then, the number of cells was compared to that of HUVEC adhering on the BP control membranes after 4 days of culture, as performed by others on bovine pericardium substrates [30,31]. Endothelial cells have been chosen because, among the different cell populations, they better mimic a physiological environment in cardiovascular applications [19].…”

“…Bovine pericardium, first utilized for bioprosthetic heart valve fabrication, has been widely used in clinical practice [ 1 , 2 ]. Decellularization is considered the critical factor that significantly promotes bovine pericardium and other tissue-derived extracellular matrix (ECM) for biomedical research and application.…”

“…The predominant cell is the pericardial fibroblast. The serous pericardium is composed of mesothelium (epithelial-like cells) with its basal lamina overlying a thin layer of loose connective tissue ( 25 ). The mesothelial cells form a monolayer lining in the visceral pericardium ( 26 ), which plays an important role in inflammation and tissue repair ( 27 ).…”
