Bovine Serum Albumin Fluorescence Quenching by Tannic Acid in Dimethylsulfoxide Containing Solutions

Abstract: Bovine serum albumin (BSA) interaction with tannic acid (TA) has been studied in dimethylsulfoxide (DMSO) aqueous solutions at different temperatures (293 and 303 K). To find out the fluorescence quenching mechanism of BSA in the presence of TA, the fluorescence data were analyzed according to the modified Stern-Volmer equation based on the approach of the existence of a “sphere of action” (a type of apparent static quenching). The values of apparent static and bimolecular quenching constants were calculated. … Show more

“…The interaction of a compound with a fluorophore can affect the fluorescence characteristics of the fluorophore due to ground-state complex formation, excited-state reactions, fluorescence resonance energy transfer (Förster's theory) or molecular rearrangements [9][10][11][12]. Fluorescence spectra of BSA in the presence of 4H2MQ are presented in Fig.…”

“…We used urea as a mild chaotropic agent to increase the availability of albumin functional groups for thiol-disulfide exchange. Chaotropic effects begin to appear after 2-3 h, so in order to speed up the reaction and provide a higher activation energy between the protein and the chaotropic agent, we used ultrasound treatment [35].…”

“…It dramatically changes the physicochemical characteristics (dielectric constant, refractive index, electrical conductivity, density, viscosity etc.) of binary solvents [7,8], thus affecting the fluorescence decay processes [9,10]. It can promote the formation of more rigid, conjugated molecular structures thus affecting the fluorescence quantum yield (Φf) of chromophore.…”
