Box-Wilson Design for Optimization of in vitro Levan Production and Levan Application as Antioxidant and Antibacterial Agents

Abstract: Background: Levan or fructan, a polysaccharide of fructose, is widely used in various commercial industries. Levan could be produced by many organisms, including plants and bacteria. The cloning of the gene from Bacillus licheniformis, which expressed levansucrase in Escherichia coli host, was carried out successfully. In the present study, we performed the in vitro production of levan and analyzed its potential application as antibacterial and antioxidant agents. Methods: In vitro levan production catalyzed b… Show more

“…The antioxidant property presented by exopolysaccharides depends on their structural factors, such as their molecular weight, monosaccharide content and the configuration of glycosidic bonds [19]. The antioxidant activity of LEV may be related to the presence of many hydroxyl groups in its structure, which can react with free radicals and generate chain reactions [79,80]. In our study, the antioxidant activity presented by LEV was medium and concentration-dependent.…”

“…Levan is one of the most promising microbial EPSs for a wide range of biotechnological applications due to its beneficial functional properties such as anticancer [ 215 , 216 , 217 ], antioxidant [ 218 ], antibacterial [ 215 , 218 ], anti-inflammatory, immunomodulatory [ 219 ], and prebiotic activities [ 220 ], as well as unique physicochemical properties such as high adhesive strength, low intrinsic viscosity, high water solubility, film-forming ability, heat stability, and high biocompatibility [ 221 , 222 , 223 ]. Levan is a neutral homopolysaccharide composed of fructose units connected by β-2,6-glycoside bonds in the backbone and β-2,1 in its branches.…”
