Brain-Derived Neurotrophic Factor/TrkB Signaling in Memory Processes

Abstract: Activity-dependent changes in synaptic strength are considered mechanisms underlying learning and memory. Brain-derived neurotrophic factor (BDNF) plays an important role in activity-dependent synaptic plasticity such as long-term potentiation. Recent experimental evidence supports the role of BDNF in memory processes: Memory acquisition and consolidation are associated with an increase in BDNF mRNA expression and the activation of its receptor TrkB. Genetic as well as pharmacologic deprivation of BDNF or TrkB… Show more

“…Treatment with U0126 (10 lmol/L) prevented E2-mediated increase in pNR2B (Tukey's post hoc test, p < 0.05, n = 3), while treatment with Wortmannin did not (Tukey's post hoc test, p < 0.05, n = 3), suggesting that activation of the ERK pathway but not the PI3-K pathway is involved in NR2B phosphorylation. It has previously been reported that interactions between NR2B and Src kinases are accompanied by ERK and Akt phosphorylation (Yamada and Nabeshima 2003), while others have indicated that NR2B could be directly phosphorylated by c-Src (Cheung and Gurd 2001). Whether E2-mediated phosphorylation of NR2B is dependent on Src/NR2B interaction in synaptoneurosomes will be addressed in future studies.…”

“…BDNF plays a critical role in synaptic plasticity, memory processes and storage of long-term memory (Bekinschtein et al, 2008a;Bekinschtein et al, 2008b;Yamada and Nabeshima, 2003). BDNF has also been found to enhance neurogenesis (Binder and Scharfman, 2004), thought to be important for hippocampal-dependent learning and memory.…”
