Brain nitric oxide synthases in the interleukin-1β-induced activation of hypothalamic-pituitary-adrenal axis

Abstract: The present study suggests that the IL-1β-induced transient stimulation of HPA axis activity is parallel in time and magnitude to the respective changes of nNOS in hypothalamus and prefrontal cortex, the brain structures involved in regulation of HPA axis activity.

“…As intraperitoneal administration of IL-1b activates the HPA axis as well (e.g. [17,18]), in the present study we set out to investigate whether the effects of repeated restraint stress on the excitatory synaptic transmission and long-term plasticity could be mimicked by exogenous IL-1b. Next, we tested whether intraperitoneal administration of anti-IL-1b antibody would abolish the effects of repeated restraint stress on cortical synaptic transmission and plasticity.…”

“…Probably, tadalafil affected ACTH secretion by inhibiting the PDE5 expressed in CNS (32), thus enhancing cGMP-mediated effects of NO on the hypothalamus-pituitary axis. In fact, NO-producing neurons are densely localized in PVN, and NO modulates the HPA axis by acting on different neuroendocrine pathways (5,14,24,30,33,45,46). Our hypothesis is supported by data from animals showing that: 1) NO exerts a tonic negative influence on the HPA axis (16), 2) NO may differently influence (e.g., increase or decrease) the activation of PVN during stress (24,27,51), and 3) in Yucatan swine, the inhibition of NO synthase increased the ACTH response to exercise (24).…”

“…The prefrontal region of the brain is involved in planning, coordinating executive decisionmaking capacity, and regulating emotional and sentient experiences. Prefrontal cortex also regulates the hypothalamic-pituitary-adrenal axis of the stress response pathways [145].If the salience is less relevant to the organism's biology as an integrated dynamical system, the endogenous protective response may be more local and limited in scope. Non-salient NPs at low pulsed doses would have minimal transient effects either in terms of overall toxicity or therapeutic effects at the level of organizational scale of the organism.…”
