Brain Tumors in Owl Monkeys Inoculated with a Human Polyomavirus (JC Virus)

Abstract: Owl monkeys were inoculated intracerebrally, subcutaneously, and intravenously with JC, BK, or SV40 virus. Two of four adult owl monkeys inoculated with JC virus, a human polyomavirus, developed brain tumors at 16 and 25 months after inoculation, respectively. A grade 3 to grade 4 astrocytoma (resembling a human glioblastoma multiforme) was found in the left cerebral hemisphere and brainstem of one monkey. The second monkey developed a malignant tumor in the left cerebral hemisphere containing both glial and n… Show more

“…The majority of the monkeys were inoculated intracerebrally as adults, and a small subset received immunosuppressive drugs prior to and following inoculation to aid in tumor inducement (London et al, 1978). Several of the infected animals, with and without immunosuppression, developed glial neoplasias in adulthood, including glioblastoma multiforme and astrocytoma.…”

“…Particular isolates of JCV, administered by various routes into hamsters and monkeys, induced the formation of neuroectodermal tumors [56][57][58][59][60][61]. A viral preparation from what was described as a Mad-1-induced owl monkey astrocytoma was injected into a second owl monkey (OWL586).…”

“…HJC-15 cells (London et al, 1978) were maintained in Dulbecco's modi®ed Eagle's medium supplemented with 10% fetal calf serum (FCS), 100 units/ml of penicillin and 100 mg/ml of streptomycin. The plasmid pTetTAk was transfected into 10 cm plates of HJL-15 cells via calcium phosphate precipitation (Graham and van der Eb, 1973).…”
