BRCA2 is a mediator of RAD51‐ and DMC1‐facilitated homologous recombination inArabidopsis thaliana

Abstract: Summary• Mutations in the breast cancer susceptibility gene 2 (BRCA2) are correlated with hereditary breast cancer in humans. Studies have revealed that mammalian BRCA2 plays crucial roles in DNA repair. Therefore, we wished to define the role of the BRCA2 homologs in Arabidopsis in detail.• As Arabidopsis contains two functional BRCA2 homologs, an Atbrca2 double mutant was generated and analyzed with respect to hypersensitivity to genotoxic agents and recombination frequencies. Cytological studies addressing … Show more

“…The primary antibodies used in this study were anti-ASY1 , anti-ZYP1 (Higgins et al, 2005), anti-RAD51 (Mercier et al, 2003), and anti-MUS81 (Higgins et al, 2008a). In the case of simultaneous staining of proteins with primary antibodies from the same species, the first primary antibody was covered with labeled Fab fragments before application of the second primary antibody (Seeliger et al, 2012). Analysis of chiasmata and statistical analysis was performed as described (Sanchez-Moran et al, 2002;Higgins et al, 2004).…”

“…Following the formation of DSBs by Spo11 (for sporulation11) (Keeney et al, 1997;Hartung and Puchta, 2000;Grelon et al, 2001;Stacey et al, 2006;Hartung et al, 2007b), recombination is initiated between homologous chromosomes by the recombinases Rad51 (for radiation sensitive51) and Dmc1 (for disrupted meiotic cDNA1) (Klimyuk and Jones, 1997;Li et al, 2004;Shinohara and Shinohara, 2004;Seeliger et al, 2012). These proteins facilitate the invasion of single-stranded ends of a DSB into homologous sequences, creating a D-loop structure.…”

“…Importantly, loss of Brca2 in plants causes chromosomal aberrations during meiosis (14). In humans, GST-pull down assays using peptide fragments of BRCA2 mapped a unique DMC1 interacting site to residues 2386-2411 (8).…”

“…In Arabidopsis, defects detected by the SHR assay could be correlated with meiotic defects in RAD51C and BRCA2 mutants (Abe et al, 2005;Seeliger et al, 2012). In addition, the use of SHR lines also permitted the identification of proteins involved in processing of recombination intermediates or in postreplicative repair (Mannuss et al, 2010).…”
