Breast Tumor Targeting with PAMAM-PEG-5FU-99mTc As a New Therapeutic Nanocomplex: In In-vitro and In-vivo studies

The IC50 dose (a dose that kills 50% of cells) was obtained for FA-CS-miR-126 nano-complex for a concentration of 500 nM at 48 and 72 h of post-treatment.…”

“…For FA-CS nano-complex, MRC5 cell viability values were respectively obtained 90.94 ± 4.79%, 92.76 ± 4.82%, and 85.45 ± 4.36% for 125 nM at 24, 48, and 72 h of post-treatment whereas these amounts were 83.60 ± 5.02%, 86.94 ± 4.78%, and 80.68 ± 3.86% at the same conditions which are indicating FA receptormediated internalization of FA-CS in cancer cells since this receptor has been significantly overexpressing on the surface of cancer cells which, in turn, improve high values of cellular uptake and result in the high level of accumulation and cytotoxicity. 17,18,20 Furthermore, CS-miR-126 nano-complex, as non-target therapeutic nanocomposite, exhibited high cell toxicity at a concentration of 500 nM after 72 h of treatment that was 69.32 ± 4.38% and 64.76 ± 3.99% for MRC5 and A549 cell lines, respectively (Figure 4(c) and (f)).
