Bright field microscopic cells counting method for BEVS using nonlinear convergence index sliding band filter

Abstract: BackgroundThe Baculovirus Expression Vector System (BEVS) is a very popular expression vector system in gene engineering. An effective host cell line cultivation protocol can facilitate the baculovirus preparation and following experiments. However, the counting of the number of host cells in the protocol is usually performed by manual observation with microscopy, which is time consuming and labor intensive work, and prone to errors for one person or between different individuals. This study aims at giving a b… Show more

“…Computer-assisted cell counting using image processing and analysis has been continuously developed and improved to provide the requirements of high reliability, specificity and sensitivity for different cell types and test protocols. For example, Sui et al [12] developed a cell counting method for host cells in a bright field for insect cells by using a nonlinear transformed sliding band filter. They obtained a low error rate and a high accuracy when comparing their method with a manual counting.…”

“…We evaluate the cell detection and counting results by using the traditional protocol employed in [4], [23], [47], [48]. We compare the results from the proposed LSTM-CFCN framework and other state of the art methods.…”

“…According to the literature review, a hemocytometer is a typical counting cell that has been used to count algal cells under a microscope (Karlson et al, 2010). A hemocytometer slide with nine big squares, from which the middle square with 400 grids is considered for counting the algal cells (Sui et al, 2014a). First, the surface of the hemocytometer was disinfected, and then ten μL of the algal solution was added to the hemocytometer according to the method described by Sui et al (Sui et al, 2014a).…”
