Monepantel resistance was diagnosed during routine monitoring of the effectiveness of a farm's roundworm control strategy. Weaned lambs had become ill thrifty and developed diarrhoea, despite the routine use of monepantel. This clinical presentation was caused by trichostrongylosis. The faecal egg count reduction was 76.7% (95% CI: 55.1-82.2%) following treatment with 2.5 mg/kg monepantel. Predominantly Trichostrongylus vitrinus along with small proportions of Oesophagostomum venulosum and Trichostrongylus vitrinus were identified by deep amplicon sequencing of pools of larvae recovered from pre and post monepantel treatment coprocultures and on postmortem examinations. The undifferentiated FECRT showed resistance to monepantel, but not to levamisole, ivermectin, or moxidectin. Examination of farm anthelmintic treatment and animal movement records suggested that treatments before movement onto silage aftermaths, putatively with low numbers of susceptible nematodes in refugia, may have placed a high selection pressure on monepantel resistance. Effective control of parasitic gastroenteritis using anthelmintic drugs is a prerequisite for sustainable sheep production. This case reiterates the need for care when combining anthelmintic treatments with movements to safe grazing, and the value of monitoring of anthelmintic efficacy as part of iterative planned animal health management.

The objective of this study is to evaluate the effect of dry wormwood and mallow on the gastrointestinal parasite of small ruminants Haemonchus contortus. Twenty-four experimentally infected lambs were randomly divided into four groups of six animals each: unsupplemented lambs, lambs supplemented with wormwood, lambs supplemented with mallow and animals supplemented with a mix of both plants. Faecal samples from the lambs were collected on day 23, 29, 36, 43, 50, 57, 64 and 75 post-infection for quantification of the number of eggs per gram (EPG). The mix of both plants contained phenolic acids (10.7 g/kg DM) and flavonoids (5.51 g/kg DM). The nematode eggs were collected and in vitro egg hatch test was performed. The aqueous extracts of both plants exhibited strong ovicidal effect on H. contortus, with ED50 and ED99 values of 1.40 and 3.76 mg/mL and 2.17 and 5.89 mg/mL, respectively, in the in vitro tests. Despite the great individual differences between the treated lambs in eggs reduction, the mean EPG of the untreated and treated groups did not differ (p > 0.05). Our results indicate that using wormwood and mallow as dietary supplements do not have a sufficient effect on lambs infected with H. contortus.

El objetivo de este estudio fue evaluar la eficacia del monepantel y levamisol sobre los helmintos gastrointestinales en rebaños ovinos del oeste de Santa Catarina. El estudio fue realizado en dos etapas. En la primera de ellas se seleccionaron cinco granjas y se formaron tres grupos (K, L y M) de 8 ovinos cada uno. En los animales del grupo K se administró monepantel (2,5 mg/kg), en los del grupo L, levamisol (5,0 mg/kg) y los animales del grupo M se utilizaron como control (no medicados). Las muestras de heces fueron recogidas el día 0 y 10 de experimento, analizadas por la técnica de McMaster com el objetivo de determinar el número de huevos por gramo de heces (HPG), seguido de la técnica de coprocultivo para la identificación de los parásitos. La segunda etapa del estudio fue desarrollada en una granjas, donde se formaron los mismos tres grupos descritos anteriormente con sus respectivos tratamientos, y cinco días después de la administración del antiparasitario se realizó el muestreo de heces de estos animales para la realización de las pruebas ecotoxicológicas, utilizando colémbolos (Folsomia candida) como marcador biológico. Para simular el uso de las heces de ovinos como abono orgánico, definimos las dosis 0; 2; 4; y 8 toneladas de heces por hectárea, así como las pruebas de supervivencia y reproducción de acuerdo con las metodologías ISO.
