Brominated Carbazole with Antibiotic Adjuvant Activity Displays Pleiotropic Effects in MRSA’s Transcriptome

Abstract: Methicillin-resistant Staphylococcus aureus (MRSA) is a major threat to human health, as the US mortality rate outweighs those from HIV, tuberculosis, and viral hepatitis combined. In the wake of the COVID-19 pandemic, antibiotic-resistant bacterial infections acquired during hospital stays have increased. Antibiotic adjuvants are a key strategy to combat these bacteria. We have evaluated several small molecule antibiotic adjuvants that have strong potentiation with β-lactam antibiotics and are likely inhibiti… Show more

“…Therefore, we treated this strain with the same conditions for RNA-seq experiments. This approach has recently been successful in uncovering transcriptome-wide alterations in gene expression due to antibiotic adjuvant compounds used alone and in combination with antibiotics . Our experimental design allowed us to measure differentially expressed genes (DEGs) that occur when S.…”

“…This approach has recently been successful in uncovering transcriptome-wide alterations in gene expression due to antibiotic adjuvant compounds used alone and in combination with antibiotics. 15 Our experimental design allowed us to measure differentially expressed genes (DEGs) that occur when S. aureus cultures were treated with loratadine, oxacillin, or both in combination ( Figure 1 A).We primarily focused on two pairwise comparisons: (1) loratadine compared to untreated and (2) cotreated compared to oxacillin treated cells. We will refer to these comparisons as Lor vs Un and Co vs Ox.…”
