Bruton’s Tyrosine Kinase Is Involved in miR-346-Related Regulation of IL-18 Release by Lipopolysaccharide-Activated Rheumatoid Fibroblast-Like Synoviocytes

Abstract: MicroRNAs (miRNAs) have emerged as key players in the regulation of expression of target mRNAs expression. They have been associated with diverse biological processes, and recent studies have demonstrated that miRNAs play a role in inflammatory responses. We reported previously that LPS-activated fibroblast-like synoviocytes (FLS) isolated from rheumatoid arthritis (RA) patients express IL-18 mRNA but they do not release IL-18. Based on the observation that this inhibition was due to a rapid degradation of IL-… Show more

“…81 miR-346 is strongly induced by LPS treatment and targets Btk mRNA in the synovial fibroblasts of rheumatoid arthritis patients. 82 Whether miR-346 targets Btk in macrophages needs to be further studied. Finally, IKKa, one of the IkB kinases in the canonical NFkB pathway, has also been confirmed as a miR-223 targets.…”

“…miRNAs are involved in diverse biological processes, including development, cancer, autoimmunity, and so on (15)(16)(17). Dysregulation of miRNAs by several mechanisms has also been described in various disease states, including SLE (18)(19)(20). However, to the best of our knowledge, studies relevant to miRNAs and lupus CD4 + T cell DNA hypomethylation are still lacking.…”

“…In particular, miR-223 is up-regulated in CD4+ naïve T lymphocytes of RA patients, and a possible role of this miRNA in the pathogenesis of the disease has been hypothesized [59]. Alsaleh and colleagues [60] studied the overexpression of miR-346 in RA fibroblast-like synoviocytes, showing that miR-346 indirectly regulates the release of the pro-inflammatory cytokine IL-18.…”
