Bta-miR-10b Secreted by Bovine Embryos Negatively Impacts Preimplantation Embryo Quality

Abstract: In a previous study, we found miR-10b to be more abundant in a conditioned culture medium of degenerate embryos compared to that of blastocysts. Here, we show that miR-10b mimics added to the culture medium can be taken up by embryos. This uptake results in an increase in embryonic cell apoptosis and aberrant expression of DNA methyltransferases ( DNMTs ). Using several algorithms, Homeobox A1 ( HOXA1 ) was identified as one of the potential miR-10b target genes an… Show more

“…Embryos secrete miRNAs that participate in embryo–maternal communication. The miRNA secretion varies according to embryo kinetic or developmental competence [ 12 , 27 , 28 ]. The identification of specific miRNAs related to embryo quality may be a non-invasive method that improves the selection of embryos with higher chance of implantation, hence the success of bovine breeding programs.…”

“…Almost 80% of identified miRNAs were also detected in EVs secreted during bovine embryo blastulation (days 5–7.5) [ 12 ]. Moreover, detected miRNAs such as bta-miR-1, bta-miR-10b, and bta-miR-184 have been detected in culture medium of bovine embryos and related to embryo competence [ 12 , 27 , 28 ].…”

“…Biomarkers have the potential to be useful in dementia in several ways, including distinguishing different aspects of underlying pathology, detection of pre-symptomatic pathological changes, predicting decline or conversion between clinical disease states, and monitoring disease progression and response to treatment ( 32 ). As mentioned, the diagnosis of FTD is particularly challenging because the relationship between clinical symptoms, pathology, and genetic causes are complex ( 31 , 55 , 56 ). In this scenario, biomarkers represent a potentially informative diagnostic tool for this condition.…”

“…Loss of short- and long-term memory and cognitive deficits are the classical characteristics of AD, but some AD patients also present with pronounced behavioral changes reminiscent of FTD [ 62 ], highlighting the heterogeneity in a single phenotype [ 63 ]. Parkinsonism can be present in both AD and FTD patients [ 26 , 27 ].…”
