Butyrate mediates nucleotide‐binding and oligomerisation domain (NOD) 2‐dependent mucosal immune responses against peptidoglycan

Abstract: The interaction between digestive tract microbiological flora and food has an important influence on human health. Butyrate is produced during the fermentation of dietary fibres by intestinal bacteria and plays an important role in the regulation of mucosal immunity. In this report, we studied the impact of butyrate on the defence mechanism against the bacterial membrane component peptidoglycan (PGN). Butyrate was found to enhance PGN-mediated IL-8 and GRO-a production. The expression of these chemokines requi… Show more

“…Butyrate, in particular, inhibits LPSinduced migration of macrophages via inhibition of histone deacetylase activity within these antigen-presenting cells [72]. Butyrate also modulates the synthesis and secretion of IL-8, MCP-1, and CXCL-1 by intestinal epithelial cells in response to peptidoglycan lipopolysaccharides and proinflammatory cytokines [103,104]. Butyrate and propionate ameliorate the LPS-stimulated synthesis of CXCL-2 by neutrophils [98], while butyrate alone inhibits interferon (IFN)-γ-inducible protein-10 (IP-10).…”

“…It is a major cell wall component of bacteria (Kurata, 2014), and has a slight difference in the molecular structure by bacterial species (Mychajlonka et al, 1980;Moynihan et al, 2014). MurNAc linked short peptide binds and stimulates nucleotide-binding oligomerization domain (NOD) 1 and 2 (Dziarski and Gupta, 2005;Leung et al, 2009). PGN of Staphylococcus aureus and Bacillus subtilis stimulates immune response via NOD whereas PGN of Lactobacillus species did not show such effect (Volz et al, 2010;Wu et al, 2015).…”

“…Recent animal studies confirm that the expression of PRRs in epithelial and immune cells can be modified by prebiotics and by butyrate in vitro [163,164]. However, the molecular mechanisms remain to be elucidated.…”
