BY‐2 Cells: Culture and Transformation for Live Cell Imaging

Abstract: Tobacco Bright Yellow‐2 (BY‐2) suspension cells are a widely used biological material for studying plant cell morphology and physiology. These cells are easy to transform and maintain in culture and tolerate transformation with fluorescent proteins such as the green fluorescent protein and its derivatives. These, by the addition of plant or mammalian targeting sequences, can be directed to specific subcellular locations for the study of cell dynamics in vivo. This unit describes the production of BY‐2 cell sta… Show more

“…Shown are representative epifluorescence micrographs of tobacco (Nicotiana tabacum) Bright Yellow-2 (BY-2) suspensioncultured cells, which serve as a well-characterized system for studying protein localization in plant cells. 18 BY-2 cells were transiently transformed via biolistic bombardment with plasmid Dna encoding full-length guayule SrPP 12 (GenBank number aF541942) C-terminally fused to the n terminus of the green fluorescent protein (SrPP-GFP). Following bombardment, cells were incubated in linoleic acid, which induces a dramatic increase in the number and size of lipid droplets in these cells (see ref.…”

“…21,22 Overall, the data show that plant cells, like mammalian cells, but unlike Saccharomyces, do not accumulate tRNAs derived from intronless and intron-containing precursors in their nucleus during nutrient deprivation, indicating that nutrient-related regulation of nuclear tRNA export is not entirely conserved among evolutionarily diverse organisms.…”

“…Given the well-documented role of Tsg101 (the mammalian homolog of Vps23) in HIV budding from the plasma membrane in mammalian cells (21,22) and that yeast Vps23p is redirected to peroxisomes in yeast cells coexpressing TBSV p33 (23), we tested whether, in an analogous manner, CIRV causes the relocalization of Vps23 to mitochondria in plant cells. Toward this end, N. tabacum BY-2 suspension-cultured cells-which serve as a well-characterized system for studying protein targeting in plant cells (57), including viral proteins (58, 59)-were transiently (co)transformed (via biolistic bombardment) with plasmids encoding fulllength CIRV and/or an N-terminal Myc epitope-tagged version of Arabidopsis Vps23 (Myc-Vps23) and then processed for immunoepifluorescence microscopy.…”
