C-C motif chemokine receptor 1 (CCR1) is a target of the EGF-AKT-mTOR-STAT3 signaling axis in breast cancer cells

Abstract: The CC motif chemokine receptor 1 (CCR1) has been implicated in tumor invasion and metastasis in numerous cancers. However, the detailed mechanism of CCR1 upregulation in metastatic tumor cells is poorly understood. The aim of this study was to clarify the regulatory mechanism underlying transcriptional activation of the CCR1 gene in response to epidermal growth factor (EGF) stimulation in breast cancer cells. CCR1 was highly expressed in human breast invasive ductal carcinoma (IDC) compared to adjacent normal… Show more

“…5257 CCR1 and its ligand CCL5 have been associated with cancer cell invasion and metastasis. 5860 Similar observations have been reported for the CCR2-CCL2, CCR5-CCL2,3,4,5, CCR7-CCL19,21 and CCR8-CCL18 axes. 6169 The role of FPR1 in tumor progression remains controversial as it has been associated with tumor invasion in colorectal cancer but with tumor suppression in gastric cancer.…”

“…In a separate study, CCR1 sustained liver metastasis by promoting local recruitment of bone marrow (BM)-derived cells that secrete the MMP9 and MMP2 metalloproteinases required for tissue invasion 45 . Activation of EGFR signaling via CCR1 has been shown to contribute to breast cancer invasion and metastasis 46 . The CCR1 mediated interaction between macrophages and breast cancer cells induced a chemokine cascade that enhanced metastasis 19 .…”

“…19,21 . CCR1 seems to be a very important receptor because it is a target of several chemokines [CC motif chemokine ligand 2 (CCL2), CCL3, CCL4, CCL5, CCL6, CCL7 and CCL9], 4,[23][24][25][26][27] which have been suggested to be involved in the pathogenesis of neuropathic pain. [28][29][30] There is some evidence demonstrating that CCR1 is involved in nociception.…”
