C-C motif chemokine receptor 9 positive macrophages activate hepatic stellate cells and promote liver fibrosis in mice

Abstract: Chemokine receptors mediate migration of immune cells into the liver, thereby promoting liver inflammation. C-C motif chemokine receptor (CCR) 9+ macrophages are crucial in the pathogenesis of acute liver inflammation, but the role and underlying mechanisms of this macrophage subset in chronic liver injury and subsequent liver fibrosis are not fully understood. We confirmed that tumor necrosis factor alpha (TNF-α)-producing CCR9+ macrophages accumulated during the initiation of carbon tetrachloride (CCl4)-indu… Show more

“…In addition, CXCL12 (SDF-1) was recently recognized as a crucial factor balancing regenerative and fibrogenic response by sinusoidal endothelial cells, and activation of CXCR4 in sinusoidal endothelial cells was found to induce profibrogenic response in HSCs [98]. Additional chemokine receptors that modulate the profibrogenic features of HSC include CCR9, the activation of which, in response to CCL25, functions as direct stimulus for HSCs activation [99].…”

“…15 CCL25 was found to attract proinflammatory macrophages via CCR9 into the liver but may also have additional migratory and activating effects on HSCs directly. 119 The proinflammatory and profibrogenic recruitment and activation of macrophages is further amplified by interactions with NKT cells. NKT cells constantly patrol the liver, a process mediated by endothelial expression of CXCL16 and corresponding expression of CXCR6 by most hepatic NKT cells.…”

“…The proinflammatory/profibrogenic Gr1 hi (Ly6C hi ) monocytes express CCR1, CCR2, CCR6, CCR8, and CCR9 (10,25,61,78). Activation of these receptors promotes differentiation of infiltrating monocytes into proinflammatory macrophages and drives HSC activation, as shown by genetic or pharmacological interventions or adoptive transfer experiments (10,25,61,78).…”
