c-Fos mapping of brain regions activated by multi-modal and electric foot shock stress

Abstract: Real-world stressors are complex and multimodal, involving physical, psychological, and social dimensions. However, the brain networks that mediate stress responses to these stimuli need to be further studied. We used c-Fos mapping in mice to characterize brain circuits activated by exposure to a single episode of multimodal stress (MMS), and compared these to circuits activated by electric foot shocks (EFS). We focused on characterizing c-Fos activity in stress-relevant brain regions including the paraventric… Show more

“…Using c-fos as proxy for neuronal activity in abstinence The immediate early gene c-fos can be used as a crude surrogate marker of in vivo neuronal activity brought about by a discrete stimulus [63,64]. In the CDFA model, significant c-fos protein expression was detected in the dBNST and insula 15 days into abstinence without a discrete stimulus (Figs.…”

“…To prevent these responses at inappropriate times, the PVN receives robust inhibitory inputs, compromising approximately 50% of all synaptic input 47 , that largely function to curtail CRH PVN neuron activity 9,48 . During an acute stress, the synaptic input received by these neurons shifts to promote CRH PVN activity [49][50][51][52] . A crucial part of this process is the relief of CRH PVN neurons from robust inhibition.…”

“…The paraventricular nucleus of the hypothalamus contributes to stress responses induced by foot shocks (2). Foot shocks promotes escape behavior in mice, resulting in compelled acute exercise.…”

“…Physiological and psychological stressors threaten the homeostasis of the body (1). In recent years, acute unpleasant exercise (AUE) caused by foot shock has been considered as a stressor that can disturb homeostasis and affect the actions of many hormones in the body (2). This stress model has been shown to cause anemia and oxidative stress(3-5).…”
