c-myc Promoter-binding Protein 1 (MBP-1) Regulates Prostate Cancer Cell Growth by Inhibiting MAPK Pathway

Abstract: Prostate cancer is the most common and invasive type of cancer among American men, and the second leading cause of cancer-elated deaths in the United States. Unfortunately, an effective therapeutic regimen is still lacking for advance stages of the disease. Recently, MEK5 has been shown to overexpress in prostate cancer and is associated with poor survival outcome. MEK5 exists as ␣-and ␤-isoforms.

“…Preoperative prostate-specific antigen (pre-PSA) level ranged from 0.3 to 97.5 ng/mL (mean, 10.36 ng/mL), with 30% of patients having a pre-PSA level greater than 10.36 ng/mL. Lower Gleason score (GS) (3)(4)(5)(6) was present in 33% patients, whereas 67% had a higher GS (7)(8)(9)(10). Biochemical recurrence occurred in 15.5% of patients, whereas 5.6% had lymph node (LN) metastases.…”

“…This could explain why PCa can respond to initial androgen withdrawal, but many patients with PCa end up with recurrence and androgen independence, leading to accelerated disease progression and death [2,3]. A number of regulatory pathways such as androgen receptor (AR) signaling pathway [4,5], mitogen-activated protein kinase signaling pathway [6], and Akt/PKB (protein kinase B) signaling pathway [7,8] may play critical roles in prosurvival/proliferation in PCa.…”

“…In addition to c-Myc, cyclin D1 expression is also suppressed by MBP-1 (Ghosh et al, 2005a). Therefore, effects of MBP-1 on mRNA expressions of COX-2, c-Myc, and cyclin D1 were also evaluated in SC-M1/HA-MBP-1 #6 cells by real-time PCR.…”

“…The 37-kDa MBP-1 is produced by alternative translation initiation from ␣-enolase gene but without enzyme activity of enolase (Feo et al, 2000;Subramanian and Miller, 2000). So far, several MBP-1-associating proteins were identified, including histone deacetylase HDAC1 (Ghosh et al, 1999), MIP2A/sedlin (Ghosh et al, 2001), MEK5␣ (Ghosh et al, 2005a), NS1-BP (Perconti et al, 2007), and Notch1 receptor intracellular domain (Hsu et al, 2008). The downstream target genes of MBP-1 remain unclear exclusive of c-myc.…”
