c-Myc regulates expression of NKG2D ligands ULBP1/2/3 in AML and modulates their susceptibility to NK-mediated lysis

Abstract: Key Points AML cells resistant to cytarabine are more susceptible to NK-mediated cell lysis. c-Myc regulates ULBP1/2/3 expression and interferes with NK cell susceptibility in primary cytarabine resistant AML blasts.

“…7A). In contrast to our study, Nanbakhsh et al 31 observed down-regulation of NKG2D ligands on Cytarabine-resistant cell lines upon treatment with Myc inhibitors. In our study only non-Cytarabine-resistant cell lines were tested and it is possible that differences in the cell lines used might account for the deviating observations.…”

“…Hence, this is an oncogenetic alteration, probably acting through the nuclear factor kB pathway. More than 20 fusion partner genes have been describes so far, 9 as confirmed by Cheah et al 1 From a diagnostic point of view, these PDGFRB diseases present with a clinical picture of chronic myelomonocytic leukemia with eosinophilia, atypical chronic myelogenous leukemia, or myelodysplastic syndrome/ myeloproliferative neoplasm overlap disorders. Imatinib is considered a definitive treatment of PDGFR-activated diseases.…”
