Background Tricholepis roylei Hook. f. is a bushy annual shrub of the Asteraceae family and point endemic species of the Western Himalaya. T. roylei is a critically endangered species and factors like poor seed germination and habitat destruction are further rendering the species towards extinction. Therefore, the present investigation was designed to document the seed germination potential of the species and to establish a reproducible in vitro propagation and mass multiplication protocol for the ex situ conservation of T. roylei germplasm. Results Seeds of T. roylei were collected from Sangdha, Himachal Pradesh, India, and were sown in different substrates under open and controlled conditions. Though the overall seed germination potential of the species was reported to be very low and maximum 45% germination was observed in cocopeat substrate after 26 days of incubation. Half-strength Murashige and Skoog (MS) medium enriched with 6-benzylaminopurine (4.4 μM l −1 ) and naphthalene acetic acid (1.0 μM l -1 ) was observed to be the optimum medium for shoot induction in shoot tip explants of T. roylei . Maximum 98.89% shoot formation was observed with 28.42 shoots per culture and 4.4-cm shoot length, respectively. The healthy shoots (4.0 cm) were transferred onto rooting media (1/4, 1/2 and full MS) and roots were developed after 8 weeks of incubation in the half-strength MS medium. Half MS augmented with 4.9 μM l −1 indole butyric acid was observed to be optimum for the root development and an average of 10.2 roots per shoot with 4.0-cm length was obtained. Rooted plantlets were successfully acclimatized under greenhouse conditions and subsequently established in the field, with a recorded survival rate of 90%. The plants acclimatized to the open environment were also planted in the wild under the habitat rehabilitation and species recovery programme. Conclusion The seed germination study envisages that the seed germination potential of the species is very poor and might be one of the probable factors responsible for the shrinkage of T. roylei population in the wild. The standardized micropropagation protocol can be helpful for the ex situ conservation of germplasm and rehabilitation of species in the wild. Moreover, the study could be helpful in elucidating the phytochemical and molecular analysis of species.

Encompassing 1.69% land area of India, Uttarakhand State sustains more than 25% species of flowering plants of India reflecting richness of flora. Large numbers of species in the state are threatened and several sources have come up with their own lists of threatened species using different threat categories leading to ambiguity. This communication attempts to compile a complete list of threatened Angiosperm species from eleven authentic sources with updated nomenclature, systematic position, original sources, threat assessment, elevational and global distribution. A total of 290 species belonging to 176 genera, 63 families, and 29 orders are listed which represent about 6% of the total flora. Elevational distribution of species shows that the 2–3 km elevation zone harbors more than half of the threatened flora (52.14%) and more than 44% endemic species despite the fact that maximum species richness is known in the 1–2 km elevation zone. Perusal of literature shows that selection of species for micropropagation is skewed towards medicinal plants rather than only threat status of a species. A disparity exists in two important sources (IUCN Red List 2020–21 and Indian Red Data Book) listing threatened taxa with only six species common to both. Eight additional species in IUCN Red List 2020–21 and 49 additional species in Indian Red Data Book are not included and vice versa. 267 species listed as threatened in various sources are not even evaluated by recent IUCN Redlist guidelines and thus warrant their immediate assessment to understand their correct present status in nature.

Lilium longiforum Thunb. adalah florikultura potensial untuk dikembangkan di bidang industri farmasi dan florikultura. Perbanyakan L. longiflorum secara generatif sulit dilakukan dan perbanyakan vegetatif dengan kultur jaringan jauh lebih efektif. Oleh karena itu, diperlukan sebuah protokol perbanyakan L. longiflorum secara in vitro yang efisien. Tujuan penelitian ini adalah mengamati induksi kalus dari eksplan sisik umbi dari planlet L. longiflorum dan respons pertumbuhannya terhadap penambahan auksin dan sitokinin dalam media kultur. Respons sisik umbi pada induksi kalus diuji dengan dua perlakuan, yaitu MS + 3,0 mg L-1 2,4-D + 0,5 mg L-1 BAP dengan inkubasi dalam keadaan 24 jam gelap (perlakuan 1) dan MS + 1,5 mg L-1 2,4-D + 1,0 mg L-1 BAP dengan fotoperiode 16/8 jam (perlakuan 2), selama 28 minggu. Kemudian, respons regenerasi kalus menjadi tunas diuji dengan penanaman kalus pada media regenerasi (MS + 3,4 mg L-1 BAP + 0,09 mg L-1 NAA) selama 12 minggu. Hasil penelitian menunjukkan bahwa eksplan pada perlakuan 2 lebih responsif untuk menginduksi kalus dari sisik umbi L. longiflorum dibandingkan eksplan pada perlakuan 1. Kalus yang dihasilkan bertekstur kompak dan berwarna hijau kekuningan dengan tingkat kesintasan 100% dan daya proliferasi yang tinggi. Media regenerasi berhasil meregenerasikan kalus menjadi tunas sebesar 100%, meskipun tidak terdapat pertumbuhan akar dalam penelitian ini. Perlakuan MS + 1,5 mg L-1 2,4-D + 1,0 mg L-1 BAP dengan fotoperiode 16/8 jam direkomendasikan sebagai sebuah protokol yang efektif dalam pengembangan L. longiflorum.
