Candidate drug replacements for quinacrine in cutaneous lupus erythematosus

Abstract: Cutaneous lupus erythematosus (CLE) is a disfiguring and potentially disabling disease that causes significant morbidity in patients. Antimalarials are an important class of medication used to treat this disease and have been the first-line systemic therapy since the 1950s. Quinacrine, in particular, is used as an adjuvant therapy to other antimalarials for improved control of CLE. Quinacrine is currently unavailable in the USA, which has taken away an important component of the treatment regimen of patients w… Show more

“…Moreover, quinacrine caused a significant decrease in the relative kidney to rat body weight ratio (Figure 1B), serum creatinine (Figure 1C) and urea (Figure 1D) concentrations, compared with the cisplatin group. erythematosus, and cutaneous sarcoidosis [13][14][15]. Mechanistically, several studies have reported the link between quinacrine and inflammation via p53 and superoxide dismutase activation as well as NF-κB and phospholipase A2 inhibition.…”

“…Quinacrine is an FDA-approved antimalarial drug and a well-known anti-inflammatory agent. It has also been reported to be beneficial in managing cancer, lupus erythematosus, and cutaneous sarcoidosis [ 13 , 14 , 15 ]. Mechanistically, several studies have reported the link between quinacrine and inflammation via p53 and superoxide dismutase activation as well as NF-κB and phospholipase A2 inhibition.…”

“…Regarding HCQ, the algorithms suitable for Japanese or Asian people was proposed who still have some restrictions on drugs to be administered [6,33] , but there are many important issues to be clarified such as the selection of the type of skin lesions for which HCQ is effective or not [2,34,35] , effects of combination therapy [36,37] and more sensitive and early diagnostic methods for retinal degeneration [29,30,31,32] .…”

“…On the other hand, due to the limited number of treatments for CLE, several new drugs are being developed. Some recent trials are introduced [37] . BI-IB059 is an antibody that targets IFN, and Anifrolumab is also an antibody against type I interferon receptor, both of which have been excellently reported in the interim report.…”
