Capabilities of liposomes for topological transformation

Abstract: Dynamic behaviors of liposomes caused by interactions between liposomal membranes and surfactant were studied by direct realtime observation by using high-intensity dark-field microscopy. Solubilization of liposomes by surfactants is thought to be a catastrophic event akin to the explosion of soap bubbles in the air; however, the actual process has not been clarified. We studied this process experimentally and found that liposomes exposed to various surfactants exhibited unusual behavior, namely continuous shr… Show more

“…Giant liposomes can be visualized by several optical microscopic methods (9-11). However, we have used dark-field microscopy because it gives the best high-contrast images of giant liposomes compared with other methods (8,(12)(13)(14)(15).…”

“…However, the electrostatic attraction alone cannot explain the formation of discoid structures, because simple adsorption is insufficient to interrupt membrane structures, as reported for interleukin-2 and interferon-gamma (Koppenhagen et al, 1998b;Van Slooten et al, 2001;Kuboi et al, 2000;Nomura et al, 2001;Sevcsik et al, 2007Sevcsik et al, , 2008Takiguchi et al, 2002). Thus, the effect of the protein structure was examined by applying DTT-treated rh-Epo to EPG-ULVs.…”

“…Great attention has been paid to understanding the principles governing the interaction of different surfactants with lipids, starting from simple model of membranes such as liposomes and vesicles [1][2][3][4][5] to more complex systems like stratum corneum. 6 Currently the physical model relates the ability of surfactants to saturate and solubilize the bilayer, with its critical micellar concentration, cmc, and its distribution coefficients, K. 7 The most employed compounds to solubilize lipidic membranes correspond to the alkyl glucoside family, specially the octyl glucoside, 1, 8-13 a non-ionic surfactant with a relatively high critical micellar concentration,18-25.3 mM.…”
