Capillary electrophoretic resolution of the 2′-, 3′- and 5′-isomers of nucleotide monophosphates via the zinc complexes

Abstract: SummaryThe affinity ofnucleotides for zinc ion enables resolution of the 2'-, 3'-and 5'-isomers of nucleotide monophosphates. The concentration of zinc ion modifies the electrophoretic mobilities of nucleoside monophosphates differently, depending on the nature of the base and the position (2', 3' and 5') of the phosphate group on the sugar moiety. Fast co-electroosmotic separation (5 min) of twelve nucleotide was performed with an acidic citrate-phosphate buffer to which zinc ion has been added.

“…Cahours et al [6][7][8] reported the addition of inorganic cations to the buffer could improve the resolution of nucleotide mono-, di-and triphosphates in CZE separation [6][7][8]. In this work, either magnesium chloride or zinc acetate was tested to see whether the resolution of UMP isomer would be improved.…”

“…Cahours et al [8] reported that the phosphate chain of the nucleotide isomers studied has the same apparent negative charge in the weak acidic medium and migration of these nucleosides monophosphate depends upon the nature of the base on the nucleotide. Therefore, the separation of nucleotides is difficult to predict.…”

“…So, using negative-end detection mode, AMPs take a longer time to reach the detection window than that in neutrally aqueous BGE. To reduce the migration time of AMP and improve the resolutions of AMPs, boric acid was usually employed, because borate can interact with the vicinal diols of nucleotides to form anionic complexes [34][35][36][37][38] in basic pH condition. In addition, Tris, an organic cation, was usually used as a buffering group in CE [32].…”
